Аннотация: An intracellular phosphorus pool in a monoculture of the cyanobacterium Spirulina platensis was assessed using radioactive and nonradioactive phosphorus. The derived dependence of specific growth rate on the intracellular content of mineral phosphorus can be presented in the form of the Droop equation. It was found that the stage of replenishment of the intracellular phosphorus pool may affect the phosphorus turnover estimation in aquatic environments from the results of short-term measurements of phosphorus uptake.

Аннотация: The source of radioactive contamination of the Yenisei River floodplain, including contamination with transuranic elements, is the Mining-and-Chemical Combine of the Russian Ministry of Atomic Energy, which has for many years been producing weapons-grade plutonium. Transuranic elements have been detected not only in the soil and sediment of the river but also in the biomass of aquatic plants. This work is an investigation of accumulation and release of 241Am by a submerged macrophyte of the Yenisei River (Elodea canadensis) in laboratory experiments. In 2000-2003, laboratory experiments were carried out with biomass of E. canadensis Mich. and filtered river water. The samples were collected from the Yenisei River upstream of the discharge of the Combine's radioactive effluent. The experiments showed that 241Am is accumulated by Elodea biomass: the activity concentration of 241Am can reach 3280 В± 240 Bq/g, with the concentration factor for 241Am 16 600 В± 2200 l/kg. Results of chemical fractionation have proved that in the course of 241Am accumulation by Elodea biomass, 241Am tightly bound to biomass increases from 11% to 27% of the total 241Am in the biomass. Release of 241Am from the decaying Elodea biomass has been evaluated experimentally. By the end of the experiment (lasting up to 127 days), the Elodea plants had lost up to 65% of their initial 241Am activity and the rate of 241Am release into the water environment reached 23 Bq/day. Аннотация: The study was done to determine concentrations of radionuclides and heavy metals and to evaluate the frequency of chromosomal aberrations in samples of Elodea canadensis, a submerged plant, collected in different parts of the Yenisei River. Samples were collected in the area subjected to radioactive impact of theMining-and-Chemical Combine (MCC) at Zheleznogorsk and in the control areas, upstream of the MCC. The investigations showed that Elodea biomass in the area affected by MCC operation contained a long inventory of artificial radionuclides typical of the MCC discharges. Upstream of the MCC, in the control sampling areas, the sediments and the Elodea biomass contained only one artificial radionuclide - 137Cs. Thus, the exposure doses to Elodea shoots and roots upstream of the MCC are small (not more than 8 ?Gy/d) and the main contribution to them is made by natural radionuclides. At the MCC discharge site (the village of Atamanovo) and downstream of it, the total dose rate increases almost an order of magnitude, reaching its maximal values - 72 ?Gy/d for Elodea shoots and 58 ? Gy/d for roots. Cytogenetic investigations of Elodea roots showed that at the MCC discharge site (the village of Atamanovo) and downstream of it the occurrence of chromosomal aberrations in ana-telophase and metaphase cells of Elodea was considerably higher than in the control area. It is highly probable that this simultaneous dramatic increase in the total exposure rate and the occurrence of chromosomal aberrations in Elodea is associated with the radiation factor. It is suggested that Elodea is affected not only by the radiation factor but also by the chemical factor - toxicity of heavy metals. В© 2009 EDP Sciences.

Аннотация: The Yenisei River, one of the largest rivers in the world, is contaminated with artificial radionuclides released by a Russian nuclear facility producing weapon-grade plutonium, which has been in operation for many years. Examination of Yenisei River sediment samples revealed the presence of artificial radionuclides typical of radioactive discharge from the Mining-and-Chemical- Combine (MCC) nuclear facility: isotopes of europium ( 152Eu, 154Eu, and 155Eu), caesium ( 137Cs and 134Cs), 60Co, 90Sr, and transuranium elements. Maximum radionuclide concentrations in sediments remained high as far as 240 km downstream of the MCC. In sediment cores collected upstream of the MCC, ?-spectrometric measurements registered only one artificial radionuclide, 137Cs, with a maximum activity of approx. 8 Bq kg -1 dry mass. Sediments of the Yenisei River also contain natural radionuclides. Sedimentation rates in several sections of the Yenisei River were determined using, different approaches: the 210Pb dating method and the ratios of artificial radionuclides - 137Cs/ 60Co and 152Eu/ 154Eu. With increasing distance downstream of the city of Krasnoyarsk, sedimentation rates increased from 0.88 cm year -1 to 1.30-1.51 cm year -1. Copyright В© 2010 IAHS Press.

Аннотация: Phototrophic sulfur bacteria form dense accumulations in the chemocline zones of stratified lakes where light reaches the sulfide-containing layers of water. Many works are dedicated to the ecophysiology of these microorganisms in meromictic lakes. However, the role of these microorganisms in the trophic network of these ecosystems, the ways of biomass utilization, and the contribution to the turnover of biogenic elements have so far been insufficiently understood. This work deals with the analysis of many years' seasonal dynamics of the biomass of purple sulfur bacteria and the physicochemical conditions of their environment in Lake Shunet (Siberia, Khakassia, Russia), unraveling the causes of their anomalous development in the chemocline of this lake, as well as the comparative analysis of such type of ecosystems. Lake Shunet is characterized by markedly pronounced stratification and the high density of purple sulfur bacteria (PSB) in the chemocline, which is comparable to that of Lake Mahoney (Canada) where the number of PSB is the greatest among those known in the world. It was shown that, in the period 2002-2009, the total amount of bacterio-chlorophyll a in the water column of Lake Shunet increased and did not correlate with the seasonal variations in temperature and illumination in the chemocline. It was established that PSB cells in the purple layer experienced the effect of self-shading. The sedimentation rate of purple sulfur bacteria in Lake Shunet was low due to the pronounced density gradient in the chemocline zone. Thus, the high number of PSB in the chemocline was due to the combination of strong illumination, a high sulfide concentration, and a high water density gradient, which was responsible for stable stratification and contributed to the accumulation of the cells in a narrow layer. The data obtained could be useful for the paleoreconstruction of climatically deter-mined changes in the level of the lake and its periods of meromixis by the presence of carotenoids and bacte-riochlorophylls in the bottom sediments. Аннотация: Data on the seasonal changes in vertical heterogeneity of the physical-chemical and biological parameters of the thermally stratified Shira Lake ecosystem (Khakasia, Siberia) in 1996-2000 have been analyzed. The interaction mechanisms involving: (1) The plankton populations in aerobic and anaerobic zones, involving the cycling of carbon and sulphur, (2) the primary production limitation (by light and phosphorus) and inhibition (by light), and (3) the kinetic characteristics of plankton populations have been elucidated. A mathematical model of the vertical structure of the lake's plankton populations, based on the ecosystem description and on vertical turbulent diffusion of the matter, has been constructed. The green alga Dictyosphaerium tetrachotomum (Chlorophyta) and the cyanobacterium Lyngbya contorta (Cyanophyta), which dominated the phytoplankton biomass, were taken as oxygen producers. Arctodiaptomus salinus (a calanoid copepod) has been assumed as the main grazer in Shira Lake as it dominated the zooplankton biomass. Four groups of microorganisms involved in the sulphur cycle formation have been distinguished: sulphur, sulphur purple, sulphur green and SRB. H2S is oxidized to sulphate (only the green sulphur bacteria oxidize it to sulphur), and sulphate is reduced to H2S, forming neither sulphur nor its water-soluble compounds. The role of grazing, light and nutrient limitation, in forming the vertical inhomogeneities, particularly in lowering the depth of the maximal cyanobacterial biomass, has been demonstrated. When the model takes into account both light limitation and nutrient limitation of algal growth by P and consumption of algae by crustaceans: (a) in the scenario where the P is formed only by the cycling and decomposition of autochthonous organic matter, both the green algae and cyanobacteria are eliminated; (b) in the scenario involving an additional P flux in the deep water layers the peak of the cyanobacteria is at a depth of 10 m, and its amplitude is close to the one observed in the lake. The position of the peak remains stable owing to the 'double' limitation mechanism: light 'from above' and P 'from below'. Another mechanism responsible for the deep position of the peak of cyanobacteria was analyzed mathematically based on the model involving the experimentally proven assumption of the growth inhibition by light in the epilimnion and the light limitation in the hypolimnion. The main result is: the peak is positioned stable at its depth and does not change with time. The analytical and numerical calculations made for this positioning mechanism yielded the formulae relating the depth of the maximum of algal biomass, the 'width' of the peak base and the peak amplitude and a number of parameters (algae elimination, turbulent diffusion coefficient, sedimentation rate, light extinction coefficient and light intensity). The theoretical curves for the stratification of chemical and biological parameters have been brought in conformity with field observations, e.g. for the different patterns for the peaks, and the biomass maxima of cyanobacteria, purple and green sulphur bacteria, oxygen, and hydrogen sulphide. The calculations revealed that for an adequate assessment of the parameters for the hydrogen sulphide zone it is necessary to introduce flows of allochthonous organic matter. For the first time, theoretically, based on the form of the sulphur distribution curve, the allochthonous input of organic matter and the inflow of hydrogen sulphide from the bottom have been discriminated. The theoretical limit for the depth up to which the hydrogen-sulphide zone can ascend under the impact of allochthonous organic loading, has been determined.

Аннотация: A new method is proposed to evaluate the interaction coefficients in microbial communities interacting due to physical-chemical environmental factors. This method differs from the classical one suggested by E.P.Odum. Redefinition of the interaction coefficients allows the evaluation of the experimental (actual) and theoretical values of the coefficients for the hypothetical interaction layout. The interaction layout is considered to be a set of factors, the values of their transformation ratios and the form of dependence of a population's specific growth rate on these factors. A comparison of theoretical and experimental values of interaction coefficients enables us to assess the adequacy of the hypothetical interaction scheme in the microbial communities. The aim of the work is to test the suggested method on a natural algal-bacterial community of Shira Lake (Khakasia, Russia), which has been the object of detailed and concerted limnological studies and for which detailed data are available. Feedback coefficients of phyto- and bacterioplankton and the coefficients of influence of phytoplankton on bacterioplankton have been defined. Dominance of negative experimental IC values has been experimentally shown, which is indicative of the negative feedback in bacterio- and phytoplankton links of Shira Lake and of negative interpopulation (phytoplankton on bacterioplankton) interactions. The considerable differences observed between the experimental and theoretical coefficients indicates inadequacy of the assumed interaction layout of the community under study. Further investigations are needed to provide a precise sketch of the interactions.

Аннотация: Batch cultures and continuous flow cultures were used to study the growth rates of zooplankton species from Shira lake, the rotifer Brachionus plicatilis Muller and calanoid copepod Arctodiaptomus salinus Daday, which were fed on phytoplankton and bacterioplankton from the lake. Analyses of the birth and survival rates were used to demonstrate that the lake phytoplankton, consisting mostly of cyanobacteria and diatomaceous algae, is inadequotes for optimal realisation of the reproductive potential of B. plicatilis when compared with the bacterial diet. The study revealed that the kinetic growth characteristics of the two zooplankters were similar: B. plicatilis rmax, 0.120 d-1; S0, 0.253; and Ks, 0.114 mg dry mass l-1; and for A. salinus rmax, 0.129 d-1; S0, 0.240; and Ks, 0.171 mg dry mass l-1. Fluctuations in natural food concentration reduced the growth rate of both species. Even though the threshold concentration of food for B. plicatilis and A. salinus were quite similar, the copepods were less sensitive to food limitation.

Аннотация: Spatial distribution of Gammarus lacustris in Lake Shira and growth of young specimens that feed on lake biota were studied. The amphipods have been shown to inhabit the littoral, sublittoral and the upper aphytal zones of the lake on stony-sandy soil and silted sand. The young and adult individuals stay apart. The young live in submerged or semi-submerged vegetation in the littoral, the adults in the sublittoral and upper aphytal zones. Maximum density of amphipods was encountered in the areas influenced by human activity. The feeding experiments revealed that the lake plankton is a more important food source for G. lacustris than any other food species. The specific growth rate measured was 0.039 d-1, with a length increment 0.095 mm d-1.

Аннотация: Synthesis of polyhydroxyalkanoates (PHAs) by a new strain of Cupriavidus - Cupriavidus eutrophus B-10646 - was investigated under autotrophic growth conditions. Under chemostat, at the specific flow rate D=0.1h-1, on sole carbon substrate (CO2), with nitrogen, sulfur, phosphorus, potassium, and manganese used as growth limiting elements, the highest poly(3-hydroxybutyrate) [P(3HB)] yields were obtained under nitrogen deficiency. In batch autotrophic culture, in the fermenter with oxygen mass transfer coefficient 0.460h-1, P(3HB) yields reached 85% of dry cell weight (DCW) and DCW reached 50g/l. Concentrations of supplementary PHA precursor substrates (valerate, hexanoate, ?-butyrolactone) and culture conditions were varied to produce, for the first time under autotrophic growth conditions, PHA ter- and tetra-polymers with widely varying major fractions of 3-hydroxybutyrate, 4-hydroxybutyrate, 3-hydroxyvalerate, and 3-hydroxyhexanoate monomer units. Investigation of the high-purity PHA specimens showed significant differences in their physicochemical and physicomechanical properties. В© 2013 Elsevier Ltd.

Аннотация: Biodegradation of film specimens from polyhydroxyalkanoates (PHAs) of two types - poly-3-hydroxybutyrate (PHB) and poly-3-hydroxybutyrate-co-3- hydroxyvalerate (PHBV) - was analysed in different environments: tropical sea waters of the South China Sea (Nha Trang, Vietnam) and soils in the environs of Hanoi (Vietnam), Nha Trang (Vietnam) and Krasnoyarsk (Siberia, Russia). In seawater, the mass loss of the specimens of both types was almost equal. However, in tropical soils, PHB degraded quicker than PHBV. In the Siberian soil, the degradation rate of the PHBV was generally higher than that of PHBV. Analysis of molecular mass of PHA specimens showed its decreasing during biodegradation. In the tropical sea conditions, PHA degrading microorganisms were represented by bacteria of Enterobacter, Bacillus and Gracilibacillus genera. Among PHA degrading bacteria, Burkholderia, Alcaligenes, Bacillus, Mycobacterium and Streptomyces genera were identified in Vietnamese soils, and Variovorax, Stenotrophomonas, Acinetobacter, Pseudomonas, Bacillus and Xanthomonas genera in Siberian soils. Micromycetes of Gongronella, Paecilomyces, Penicillium and Trichoderma genera exhibited PHA degrading activity in Vietnamese soils, and Paecilomyces, Penicillium, Acremonium, Verticillium and Zygosporium genera - in Siberian soils. Аннотация: Biodegradability patterns of two PHAs: a polymer of 3-hydroxybutyric acid (3-PHB) and a copolymer of 3-hydroxybutyric and 3-hydroxyvaleric acids (3-PHB/3-PHV) containing 11 mol% of hydroxyvalerate, were studied in the tropical marine environment, in the South China Sea (Nha Trang, Vietnam). No significant differences have been observed between degradation rates of 3-PHB and 3-PHB/3-PHV specimens; it has been found that under study conditions, biodegradation is rather influenced by the shape of the polymer item and the preparation technique than by the chemical composition of the polymer. Biodegradation rates of polymer films in seawater have been found to be higher than those of compacted pellets. As 3-PHB and 3-PHB/3-PHV are degraded and the specimens lose their mass, molecular weight of both polymers is decreased, i.e. polymer chains get destroyed. The polydispersity index of the PHAs grows significantly. However, the degree of crystallinity of both PHAs remains unchanged, i.e. the amorphous phase and the crystalline one are equally disintegrated. PHA-degrading microorganisms were isolated using the clear-zone technique, by inoculating the isolates onto mineral agar that contained PHA as sole carbon source. Based on the 16S rRNA analysis, the PHA-degrading strains were identified as Enterobacter sp. (four strains), Bacillus sp. and Gracilibacillus sp.В© 2010 Elsevier Ltd. All rights reserved.

Аннотация: The possibility of use of polyhydroxyalkanoates (PHAs), biodegradable microbial polyesters, as a carrier for pesticides (?-hexachlorcyclohexane and lindane) for targeted and controlled delivery of these compounds to soil was investigated. The kinetics of polymer degradation and the dynamics of pesticide release from the extended-release formulations was studied. It is shown that pesticides embedded in a degradable polymer (PHA) carrier are released gradually and slowly, without surges, as the polymer is degraded by the soil micro-flora. The microbial soil component actively responded to the addition of the polymer as an additional nutrient substrate: the latter was degraded and then utilized. The rate of the pesticide release to the soil can be regulated by varying the polymer-pesticide ratio. В© 2009 Pleiades Publishing, Ltd.

Аннотация: An experimental dosage form of rubomycin is developed: the drug is incorporated in absorbable polymeric (polyhydroxybutyrate) matrix in the form of microparticles. Antitumor efficiency of this rubomycin dosage form was studied in laboratory mice with transplanted Ehrlich ascitic carcinoma. Rubomycin deposited in polymeric microparticles exhibited pronounced antitumor activity, inhibited the proliferative activity of Ehrlich ascitic carcinoma, and improved survival of mice with tumors. Аннотация: The paper deals with the study of the synthesis of 3-hydroxybutyrate (3HB) and 3-hydroxyvalerate (3HV) copolymers by the bacterium Ralstonia eutropha B-5786 grown under different carbon nutrition conditions (growth on carbon dioxide, fructose, and CO2-valerate and fructose-valerate mixtures). The parameters to be analyzed included the yield of biomass, the yield, synthesis rate, and composition of copolymers, the activity of the key enzymes of polyhydroxyalkanoate (PHA) synthesis (?-ketothiolase, acetoacetyl-CoA reductase, and PHA synthase), the maximum tolerable concentration of valerate to the bacterium, and the conditions that govern the incorporation of hydroxyvalerate to copolymers. This allowed the relationship between cultivation conditions and the proportion of monomers in the copolymers to be deduced. We were able to synthesize a range of 3HB/3HV copolymers and found that the thermal characteristics and the degree of crystallinity of these copolymers depend on the molar fraction of 3HV.

Аннотация: The biodegradability of oriented fibers made of polyhydroxybutyrate (P(3HB)) and its co-polymer with ?-hydroxyvalerate (P(3HB-co-3HV)) was investigated in buffer solutions and in biological media in vitro and in vivo. The fibers of both polymer types demonstrated resistance to hydrolytic degradation in buffer solutions at 38В°C and pH from 4.5 to 7.0 (for up to 180 days). It has been found that the biodegradation of the fibers in vitro in blood and serum and in vivo is accompanied by weight losses and minor changes in the microstructure with no significant losses in the tensile strength over a long time (up to 180 days). The biodegradation rate of the less crystalline co-polymer P(3HB-co-3HV) fibers was 1.4-2.0-times higher than that of the homopolymer P(3HB). It has also been shown that the degradation of the fibers in vivo is influenced both by tissue fluid enzymes and cells (macrophages and foreign-body giant cells). The fibers were eroded on the surface only with no gross defects and no dramatic effects on their mechanical performance. В© VSP 2005.
