Аннотация: The dynamics of accumulation of polyhydroxybutyrate (PHB) and the activities of key enzymes of PHB metabolism (?-ketothiolase, acetoacetyl-CoA reductase, PHB synthase, D-hydroxybutyrate dehydrogenase, and PHB depolymerase) in the hydrogen bacterium Ralstonia eutropha B5786 were studied under various conditions of carbon nutrition and substrate availability. The highest activities of ?-ketothiolase, acetoacetyl-CoA reductase, and PHB synthase were recorded during acceleration of PHB synthesis. The activities of enzymes catalyzing PHB depolymerization (PHB depolymerase and D-hydroxybutyrate dehydrogenase) were low, being expressed only upon stimulated endogenous PHB degradation. The change of carbon source (CO2 or fructose) did not affect the time course of the enzyme activity significantly.

Аннотация: The vertical distribution and abundance of Gammarus lacustris in the pelagic zone of two fishless meromictic lakes, L. Shira and L. Shunet, in Southern Siberia (Russia), was studied with the underwater video recording system and using vertical hauls. In both lakes, during summer stratification, Gammarus was distributed non-homogenously, with a stable peak in the metalimnion. The average depth of Gammarus population in the pelagic zone was significantly correlated with the depth of the thermocline. Gammarus abundances obtained using vertical plankton hauls with net were quite comparable with those obtained from video records. The peak abundance of Gammarus in the pelagic zone of the lakes observed with underwater video amounted up to 400 individuals m-2, while the peak animal densities in the metalimnion reached 50 ind. m-3. The data are compared with previously published abundances of Gammarus in the littoral of Lake Shira. Both littoral and pelagic can be equally important habitats for amphipods in meromictic lakes. The absence of fish in the pelagic zone, high oxygen concentration, low water temperature, increased seston concentration, elevated water density in the metalimnion and the anoxic hypolimnion can be the most probable combination of factors that are responsible for the peak of Gammarus in the metalimnion of these lakes. Аннотация: A focused He - Ne laser beam is shown to cause local blackening of semitransparent detonation nanodiamond (DND) films at incident power densities above 600 W cm(-2). Data obtained with a Raman spectrometer and low-power 632.8-nm laser source indicate that the blackening is accompanied by a decrease in broadband background luminescence and emergence of sharp Raman peaks corresponding to the structures of nanodiamond and sp(2) carbon. The feasibility of image recording on DND films by a focused He - Ne laser beam is demonstrated.

Аннотация: Application of pulse-amplitude-modulation (PAM) fluorometers for measuring slow stages of chlorophyll fluorescence induction (CFI) is considered. With an example of Triticum aestuvum L. plants grown under continuous illumination at a photon flux density of 600 μmol/(m2 s) photosynthetically active radiation (PAR), the CFI curves were analyzed with leaves of various ages as a function of actinic light intensity. The fluorometer PAM-2100 was applied for measurements of CFI curves. The characteristic peaks of CFI curves in wheat leaves were most conspicuous and had the largest amplitudes at 600–800 μmol/(m2 s) PAR, which corresponds to the middle range of actinic light intensities employed in PAM-2100 fluorometers. In plants exposed to favorable and stressful conditions, the developmental stages may proceed at different rates; thus, the comparison of fluorescence parameters for leaves of equal calendar age but having different physiological states may provide ambiguous data. Therefore, the feasibility of recording CFI curves of different types is quite important for rapid diagnostics of the age and state of plant leaves, as well as for adequate physiological conclusions. © 2016, Pleiades Publishing, Ltd.

Аннотация: In studies of fungal bioluminescence, not only fruiting bodies and spores of the fungus, but also samples of luminescent wood, leaf litter or soil may need to be used to derive pure mycelial culture. This study describes an approach to isolating the culture of luminescent fungal mycelium from samples of light-emitting wood found on Borneo Island in November-December 2013. A GelDoc XR Imaging System (Bio-Rad Laboratories, Inc., U.S.) was used for the first time to monitor luminescence and select luminous samples. This study shows that for successful isolation of the culture of luminescent mycelium out of the luminescent wood found in the forest, it is imperative to keep the samples moist (mycelium alive until there is water), while immediate and aseptic delivery of the samples to the laboratory is not a crucial condition (inner layers of wood is "sterile"). Investigation of the growth features of the isolated mycelium in various growing conditions revealed some peculiar properties of its luminescence in comparison with the known luminescent cultures of basidiomycetes. When grown on solid nutrient media, mycelium exhibits low growth rates, long-lasting luminescence (140 days or longer), and emergence and disappearance of local zones with high levels of light emission. Mycelium produced in submerged culture does not emit light, and this effect must be caused by the absence or a very low level of the luminescent reaction substrate in the biomass. The luminescence system isolated from mycelial biomass did not induce luminescent reaction in vitro upon the addition of NADPH (recording intensity is 60 100 URL/sec). We found that enzymes of the luminescence systems isolated from mycelium pellets retained their activity and catalyzed luminescent reaction when a hot extract of the luminous fungus Armillaria sp. (IBSO 2360) was added (near 1900 URL/sec). The same effect was obtained after addition of hot extracts from the fruiting bodies of nonluminous higher fungi Pholiota squarrosa, Cortinarius sp., Hypholoma capnoides and Chroogomphus rutilus (near 3500 URL/sec). The pure culture of luminescent mycelium has been registered in the Culture Collection of IBP SB RAS as IBSO 2371; now it can be used for various in vivo and in vitro studies, including identification of the fungus.

Аннотация: We studied structure, dynamics and functional (biogeochemical) activity of microbial complexes of cryogenic soils in larch forests in Central Evenkia and polygonal tundra on Samoilovskii Island, Lena Delta. We found that daily flux of methane from soil surface is 3–5 times less in forest soil than in the center of polygon in tundra. Short-term heating to 18.5–22.5 °C of permafrost-affected soil in larch forest caused sweetening of soil solution, shrinkage of eco-trophic groups of microorganisms and microbial biomass, as well as increase in greenhouse gases (CO2 and CH4) emission to the air. Notably the permafrost-affected soil on sandy deposits in tundra had highest microbial diversity of methanogenic archaea including Methanobacteriaceae, Methanomicrobiaceae, Methanosarcinaceae, Methanosaetaceae families. On the other hand only Methanosarcinacea were found in cryosols of larch forest. Both type I and type II methanotrophs were found in the forest soil, while only type II methanotrophs occurred in tundra soil. © 2017, Izdatel’stvo Nauka. All rights reserved.
