Аннотация: The experiments on accumulation of 99Tc by Elodea biomass showed that 99Tc activity concentration can reach 120 В± 6Bq/g, with the concentration factor for 99Tc 2700 В± 500 L/kg. In experiments on 99Tc release, over 504 h about 82% of the total 99Tc activity was released into the water from plant biomass; most of 99Tc was released within the first 192 h. Results of chemical fractionation of the biomass show that 99Tc contained in biomass was mainly concentrated in the exchangeable and the adsorbed fractions (83%). Thus, the data obtained using chemical fractionation of biomass confirmed the experimental data on 99Tc release, which suggested that most of the biomass-bound 99Tc was adsorbed on the biomass surface. 99Tc tightly bound to Elodea biomass (fractions of organics and mineral residue) constituted just 17% of the total 99Tc activity. Decreased illumination of Elodea shoots during the experiment did not cause any reduction in 99Tc activity concentration or concentration factor. Results of chemical fractionation of the biomass grown under lower illumination conditions show that the percentage of 99Tc tightly bound to Elodea biomass (fractions of organics and mineral residue) decreased while the 99Tc of the adsorbed fractions decreased. Our results and data reported by other authors suggest that some part of 99Tc activity can be bioavailable to living organisms and that the portion of bioavailable 99Tc can be determined by a number of factors. Аннотация: Microbiological and isotopic-geochemical investigations of the brackish meromictic lakes Shira and Shunet were performed in the steppe region of Khakasia in winter. Measurements made with a submersed sensor demonstrated that one-meter ice transmits light in a quantity sufficient for oxygenic and anoxygenic photosynthesis. As in the summer season, in the community of phototrophic bacteria found in Lake Shira, the purple sulfur bacteria Amoebobacter purpureus dominated, whereas, in Lake Shunet, the green sulfur bacteria Pelodictyon luteolum were predominant. Photosynthetic production, measured using the radioisotopic method, was several times lower than that in summer. The rates of sulfate reduction and production and oxidation of methane in the water column and bottom sediments were also lower than those recorded in summer. The process of anaerobic methane oxidation in the sediments was an exception, being more intense in winter than in summer. The data from radioisotopic measurements of the rates of microbial processes correlate well with the results of determination of the isotopic composition of organic and mineral carbon (? 13C) and hydrogen sulfide and sulfate (? 34S) and suggest considerable seasonal variations in the activity of the microbial community in the water bodies investigated.

Аннотация: Microbial polyhydroxyalkanoates (PHAs), because of their well studied complex physiology and commercial potential, are vehicles for carbon and potential storage reduction for many microbial species. Even with the wealth of studies about microbial PHAs in the scientific literature, polymer accumulation and degradation are still not comprehensively understood. Poly(3-hydroxybutyrate) (P3HB) granule formation and polymer mobility were studied here in the bacterium Ralstonia eutropha strain B5786 in autotrophic cultures. Electron microscopy studies revealed decreasing cell size concomitant with enlargement of size and number of intracellular granules, and inhibition of cell division during intracellular polymer production. Activities of key P3HB biosynthetic enzymes demonstrated correlations with each other during polymer accumulation, suggesting an intricately regulated P3HB cycle in autotrophically grown R. eutropha cells. Аннотация: The physiological, biochemical, genetic, and cultural characteristics of the glucose-utilizing mutant strain Ralstonia eutropha B8562 were investigated in comparison with the parent strain R. eutropha B5786. The morphological, cultural, and biochemical characteristics of strain R. eutropha B8562 were similar to those of strain R. eutropha B5786. Genetic analysis revealed differences between the 16S rRNA gene sequences of these strains. The growth characteristics of the mutant using glucose as the sole carbon and energy source were comparable with those of the parent strain grown on fructose. Strain B8562 was characterized by high yields of polyhydroxyalkanoate (PHA) from different carbon sources (CO 2, fructose, and glucose). In batch culture with glucose under nitrogen limitation, PHA accumulation reached 90% of dry weight. In PHA, ?-hydroxybutyrate was predominant (over 99 mol %); ?-hydroxyvalerate (0.25-0.72 mol %) and ?-hydroxyhexanoate (0.008-1.5 mol %) were present as minor components. The strain has prospects as a PHA producer on glucose-containing media.

Аннотация: The biodegradability of oriented fibers made of polyhydroxybutyrate (P(3HB)) and its co-polymer with ?-hydroxyvalerate (P(3HB-co-3HV)) was investigated in buffer solutions and in biological media in vitro and in vivo. The fibers of both polymer types demonstrated resistance to hydrolytic degradation in buffer solutions at 38В°C and pH from 4.5 to 7.0 (for up to 180 days). It has been found that the biodegradation of the fibers in vitro in blood and serum and in vivo is accompanied by weight losses and minor changes in the microstructure with no significant losses in the tensile strength over a long time (up to 180 days). The biodegradation rate of the less crystalline co-polymer P(3HB-co-3HV) fibers was 1.4-2.0-times higher than that of the homopolymer P(3HB). It has also been shown that the degradation of the fibers in vivo is influenced both by tissue fluid enzymes and cells (macrophages and foreign-body giant cells). The fibers were eroded on the surface only with no gross defects and no dramatic effects on their mechanical performance. В© VSP 2005.

Аннотация: Kinetic parameters of a culture of the hydrogen-oxidizing bacterium Ralstonia eutropha, grown on a gas substrate under the conditions favoring autotrophic biosynthesis of polyhydroxybutyrate, were studied. The following parameters, making it possible to control and optimize the process in industrial situations, were determined: specific rate of substrate consumption, physical properties of culture medium, and coefficients of heat emission and mass transfer.

Аннотация: The cyanobacterium Anabaena flos-aquae and many other phytoplankton species have a complex life cycle that includes a resting stage (akinete). We present a new agent-based (also known as individual-based) model of Anabaena that includes the formation and behavior of akinetes. The model is part of a coupled Eulerian-Lagrangian model and can reproduce the main features of the observed seasonal and interannual population dynamics in Bugach Reservoir (Siberia), including an unexpectedly large bloom in a year with low nutrient concentrations. Model analysis shows that the internal loading of phosphorus (P) due to germination from the sediment bed is ?10% of the total input. However, most of the long-term nutrient uptake for Anabaena occurs in the sediment bed, which suggests that the sediment bed is not just a convenient overwintering location but may also be the primary source of nutrients. An in silico tracing experiment showed that most water column cells (?90%) originated from cells located in the sediment bed during the preceding winter. An in silico gene knockout experiment (akinete formation is prohibited) showed that the formation of resting stages is of critical importance to the survival of the population on an annual basis. A nutrient-reduction management scenario indicates that Anabaena densities increase because they are less sensitive to water column nutrient levels (because of the sediment bed source) than other species. Аннотация: Frozen samples of common fish species, sea trout (Salmo trutta), from Norway and Siberia, herring (Clupea harengus pallasi), rock sole (Lepidopsetta bilineata) and cod (Gadus morhua maris-albi), collected from a wholesale market in Krasnoyarsk city (Siberia, Russia) were analyzed. Special attention was paid to long-chain essential polyunsaturated fatty acids: eicosapentaenoic, 20:5?3 (EPA) and docosahexaenoic, 22:6?3 (DHA). Heat-treatment (cooking and frying) did not in general significantly decrease the contents of EPA and DHA compared to raw fish species, except for a modest reduction in Norwegian trout during frying. Boiled trout appeared to be a more valuable fish dish for obtaining the officially recommended appropriate daily intake of EPA + DHA for humans. Аннотация: Contents of fatty acids in filets of unfrozen (control), boiled, fried, roasted and boiled in a small amount of water humpback salmon, collected from a wholesale market in Krasnoyarsk city (Siberia, Russia) were analyzed. Special attention was paid to essential polyunsaturated fatty acids of ?3 family: eicosapentaenoic, 20:5?3 (EPA) and docosahexaenoic, 22:6?3 (DHA). Heat treatment in general did not decrease content of EPA and DHA in humpback, except a modest reduction during frying. Cooked humpback appeared to be the valuable source of essential ?3 PUFAs, namely EPA and DHA. It was hypothesized that the absence of significant reduction of PUFAs' contents in red flesh of fishes of Salmonidae family during heat treatment may be due to a high level of natural antioxidants which formed in the course of evolution as adaptation to their ecological niche. Аннотация: In 2002, a "top-down" biomanipulation (reduction of biomass of planktivorous fish Carassius auratus) had been successfully carried out in a small reservoir of the river Bugach (Krasnoyarsk, Russia), after which the cyanobacterial blooming ceased. However, the reservoir ecosystem was absolutely free of Daphnia - the main link of trophic cascade. As supposed, the reduction of blooming was the result of suppression of a direct stimulation effect of planktivorous fish on cyanobacteria, revealed earlier in laboratory experiments. The question arose as to whether the effect of stimulation of cyanobacteria revealed in laboratory may lead to the changes in biomass of cyanobacteria in the reservoir, observed after the biomanipulation. To test this supposition, a mathematical model describing growth of cyanobacteria in the reservoir was developed. The modelling results and field data on biomass of cyanobacteria in summer closely coincided. Modelling calculations showed that direct influence of planktivorous fish could cause the second summer peak of water blooming by Microcystis. On the contrary, removal of crucian carp from the reservoir will not affect the level of water blooming caused by cyanobacteria Anabaena, as this species' growth is not stimulated by fish. Аннотация: The work presents the data on the spatial distribution of temperature anomalies in the Global Ocean, based on the long-term measurements with the AVHRR satellite equipment. It is noteworthy that such results can be only obtained by using long time series of satellite images. To obtain them by ship-borne methods is actually impossible. The proposed anomaly criterion is equal to a difference between minimum/maximum and average temperature, normed for the mean square deviation. The difference is calculated from the whole time series (with regard to a season). Based on this criterion, anomalies can be distinguished on the background of usual seasonal dynamics at various absolute values of deviations of the measured parameter. Besides, rare, single, fluctuations can be also revealed. Based on the proposed criterion, connected zones have been defined. These zones occur almost in every part of the Global Ocean. The data obtained have been analyzed in association with the CZCS (1978-1986) and SeaWiFS satellite images in order to compare them with the areas of anomalous dynamics of chlorophyll concentration and the regions quasistationary in the seasonal dynamics of chlorophyll. Anomalies in the temperature dynamics are of exceptional importance for the formation of climatic deviations; they affect the ocean-atmosphere interaction and reflect variations in the hydrological structure of the surface waters. The work presents a comparison with the dynamics of chlorophyll concentration distribution determined from the SeaWiFS data for the Atlantic Ocean. В© 2003 COSPAR. Published by Elsevier Ltd. Аннотация: The paper analyzes the stability of areas in the Global Ocean, which are quasistationary with respect to the seasonal dynamics of chlorophyll concentration. Such areas were first discovered based on the CZCS data. They occur almost in all regions of the Global Ocean. They reflect typical seasonal dynamics of phytoplankton, which depends on both large ocean currents and local hydrological instability. It is related to the displacement of frontal zones and to local hydrological anomalies. Since the completion of the CZCS work there has been a 10 years gap in monitoring chlorophyll spatial distribution dynamics in the ocean. The reason is the long absence of equipment on the orbit, which would be suitable for the measurement of chlorophyll concentration. Recently, SeaWiFS data have been accumulated and their quantity is sufficient for the crude statistical analysis of image time series for 25 years. Thus, it has become possible to obtain data on the long-term stability of quasistationary areas and, therefore, on the stability of large-scale hydrological phenomena in the Atlantic Ocean. Аннотация: Variability of chlorophyll concentration in the ocean is one of the most important components of the primary production process on the planet. To preserve the biosphere and to make appropriate use of it, it is imperative to have a deep insight into the long-term dynamics of the primary production on the planet. To investigate the dynamics of chlorophyll concentration based on satellite data, the Institute of Biophysics (Russian Academy of Sciences, Siberian Branch) has developed a geoinformation system. It was used to detect the areas in the global ocean that are quasistationary in relation to seasonal dynamics of chlorophyll concentration. Areas such as these, found in the Indian Ocean, are described in this work. These areas form the basis for the analysis of long-term dynamics of chlorophyll concentration. In these quasistationary zones systematic monitoring of phytopigment concentration is conducted by space-borne and marine craft. The work presents long-term satellite-based data on the space distribution of anomalous deviations of chlorophyll concentration in the ocean. An anomaly criterion is proposed and maps of Pacific Ocean areas with a high probability of anomalies are presented. В© 2004 Taylor and Francis Ltd.

Аннотация: The new data on anomalies in seasonal dynamics of chlorophyll concentration in the ocean have altered the approach to the analysis of long-term trends of phytopigments in the surface layer of the ocean. Zones have been found in which at various times there was a many-fold increase in chlorophyll concentration versus its usual level. These changes are local in time and space. If anomalies in the long-term tendencies in the change of phytopigment surface distribution in the ocean are ignored in the analysis, the resulting picture will inadequately reflect the real processes. The work presents the results of the analysis of long-term trends of chlorophyll concentration on the basis of the CZCS satellite data in the Atlantic Ocean. The results are given as maps representing different variants of trends. The data obtained can be used to analyze the consequences of long-term changes of climate and changes in the interaction between the atmosphere and the ocean. В©2002 COSPAR. Аннотация: The work presents long-standing CZCS-satellite-based data about spatial distribution of anomalies in chlorophyll concentration and temperature of the ocean. An anomaly criterion is proposed. Maps of Atlantic ocean areas with elevated probability of anomalies are given. Properties of anomalous zones in different parts of the Atlantic ocean are discussed. Satellite-based maps of anomaly coefficients for chlorophyll concentration and temperature in the Atlantic ocean have been shown to feature substantial differences, the zones of elevated anomaly of chlorophyll concentration and temperature - to be close only in the vicinity of the Amazon outflow into the Atlantic ocean. Along with fundamental knowledge about dynamics of biological phenomena in the ocean the methods described may be used to reveal deviations due to anthropogenic impact. The methods presented may be used to process SeaWiFS prograna data. The data derived may be used to analyze long-standing time series to estimate randomness of cunent changes. В©2002 COSPAR. Аннотация: The purpose of this work was to study the full-scale potential use of human mineralized waste (feces and urine) as a source of mineral elements for plant cultivation in a biological life support system (BLSS). Plants that are potential candidates for a photosynthesizing link were grown on a neutral solution containing human mineralized waste. Spring wheat Triticum aestivum L., peas Pisum sativum L. Ambrosia cultivar and leaf lettuce Lactuca sativa L., Vitaminny variety, were used. The plants were grown hydroponically on expanded clay aggregates in a vegetation chamber in constant environmental conditions. During plant growth, a determined amount of human mineralized waste was added daily to the nutrient solution. The nutrient solution remained unchanged throughout the vegetation period. Estimated plant requirements for macro-elements were based on a total biological productivity of 0.04 kg day-1 m-2. As the plant requirements for potassium exceeded the potassium content of human waste, a water extract of wheat straw containing the required amount of potassium was added to the nutrient solution. The Knop's solution was used in the control experiments. The experimental and control plants showed no significant differences in state or productivity of their photosynthetic apparatus. A small decrease in total productivity of the experimental plants was observed, which might result in some reduction of O{cyrillic}2 production in a BLSS. В© 2009 COSPAR.

Аннотация: The purpose of this study is to estimate the possible effect produced on plant productivity by inedible plant biomass added to soil-like substrate (SLS). Results of the experiments with radish plants grown on the SLS with inedible biomass of carrot and beet plants added in the amounts roughly equal to their yields harvested from the same area showed a significant reduction in productivity of radish plants. The yield of radish plants grown on the SLS with added radish tops was almost the same as the yield of the radish grown on the neutral substrate. Experiments with addition of dry wheat straw to the SLS and growing of wheat and radish plants on that substrate also showed that the productivity of the plants grown in that way was decreased. Attempts to negate the adverse effect of plant wastes proved that the most effective way was to mineralize the wastes using the technique of "wet incineration". В© 2008 Elsevier Ltd. Аннотация: This closing technology developed for experimental BIOS-3 facility seems to be highly expedient for providing normal life conditions in high-latitude settlements and first of all for life-support of Antarctic stations. Obvious advantages of ecological life-support system based on photosynthesis, in comparison with used now in Antarctic Continent, are: 1) highly effective cascade usage of energy for performance of all functions of life-support; 2) minimization of environmental pollution due to processing human excretions and polluted water; 3) providing crew with fresh vegetable food all-the-year-round; 4) contact with alive plants is powerful source of positive emotions and effective means of psychological relaxation; 5) significant reduction of the transport charges on delivery of foodstuff to Antarctic Continent. First of all, the usage of closed ecosystem can be recommended for high-mountainous station "Concordia". Similar ecosystems can be used in northern high-latitude settlements - Russian North, Canadian Northern territories, Alaska. Finally, the experience of closed ecological life-support system operation in Antarctic Continent can serve as the best range for modeling some aspects of Martian mission - its ecological, biotechnological and psychophysiological problems - since there are no natural conditions on the Earth closer to Mars with respect to severity of a place.

Аннотация: The biogeochemical and molecular biological study of the chemocline and sediments of saline meromictic lakes Shira and Shunet (Khakasia, Russia) was performed. A marked increase in the rates of sulfate reduction and methanogenesis was revealed at the medium depths of the chemocline. The rates of these processes in the bottom sediments decreased with depth. The numbers of the members of domains Bacteria, Archaea, and of sulfate-reducing bacteria (SRB) were determined by fluorescence in situ hybridization with rRNA specific oligonucleotide probes labeled with horseradish peroxidase and subsequent tyramide signal amplification. In the chemocline, both the total microbial numbers and those of Bacteria were shown to increase with depth. The archaea and SRB were present in almost equal numbers. In the lake sediments, a drastic decrease in microbial numbers with depth was revealed. SRB were found to prevail in the upper sediment layer and archaea in the lower one. This finding correlated with the measured rates of sulfate reduction and methanogenesis. В© Nauka/Interperiodica 2006.
