Аннотация: The emission of different, diamond coatings on silicon field emitter. arrays were investigated: chemical vapor deposition diamond, high pressure and high temperature synthetic diamond, and shock-synthesized nanodiamond. Practical emission characteristics such as: emission threshold, maximum current, current stability, and reproducibility were tested. Аннотация: Solid-phase bioluminescent immunoassay of thyroid hormones, human thyrotropin (hTSH) and two forms of thyroxine (T4), whose determinations are vitally important for diagnostics of thyroid diseases and the efficiency of treatment, is described. The recombinant obelin, a Ca2+-regulated photoprotein originally derived from the luminous marine hydroid Obelia longissima, is employed as a bioluminescent label. To produce obelin conjugates with anti-hTSH, anti-T4 immunoglobulins (IgG), and T4, additional SH groups are introduced into the obelin molecule using Traut's reagent (2-iminothiolane) and then obelin possessing extra SH groups is conjugated with succinimidyl 4-(N-maleimidomethyl)-cyclohexane-1-carboxylate-activated IgGs or T4. The total yield of obelin conjugates determined by luminescent activity is 60-65% after all chemical and purification procedures. The obtained conjugates are stable to lyophilization and in solution for at least 9 months at 4 degreesC, with loss of activity not exceeding 10%. The application of obelin conjugates for determination of the hTSH, total T4, and free T4 in standard, control, and patient sera displays high sensitivity and reproducibility of results. The results of bioluminescent immunoassays are closely comparable to those obtained by the radioimmunoassay method (R = 0.95-0.99). (C) 2003 Elsevier Inc. All rights reserved.

Аннотация: The possibility of creating new screening methods with a cell-free translation system has been demonstrated with a quantitative determination of diphtheria toxin and some antibiotics (puromycin, kanamycin and tetracycline) as examples. The approach proposed follows from the ability of various substances to inhibit protein synthesis. We used a wheat-germ cell-free translation system stabilized by freeze-drying in the presence of trehalose with the mRNA of the Ca2+-activated photoprotein obelin as a reporter template. This freeze-dried cell-free translation system allows prolonged storage of the detecting system before it is required, increases the reproducibility of the results and simplifies the application procedure. The obelin mRNA extends the sensitivity of the method owing to the high sensitivity of detection of the synthesized protein.

Аннотация: Novel dual-analyte single-well bioluminescence immunoassay (BLIA) for total and IgG-bound prolactins was developed on the base of Ca2+-regulated photoprotein obelin mutants with altered color and kinetics of bioluminescence signal as reporters. The mutants W92F-H22E and Y138F were chemically conjugated with monoclonal mouse anti-hPRL and anti-hIgG immunoglobulins and thus displayed signals from total prolactin and IgG-bounded prolactin (macroprolactin) correspondingly. Bioluminescence of the reporters was simultaneously triggered by a single injection of Ca2+ solution and discriminated via bioluminescent signal spectral and time resolution. The developed microplate-based immunoassay allows detection of two prolactin forms in crude serum without additional manipulations (e.g., gel chromatography or PEG-precipitation). Total prolactin bioluminescence immunoassay in standard, control, and clinical sera offers high sensitivity and reproducibility. The BLIA results show good correlation with those obtained by RIA and immunoassay after gel chromatography.

Аннотация: The term Closed Ecological System (CES) is in wide use. However there is no generally accepted measure of the closure of ecological systems. In order to obtain reproducibility of experiments with natural and man-made CES (with respect to degree of closure) some universal estimate needs to be developed. Understanding ecological systems as a network and closure as the degree of matter recycling allows the use of matrix graphs. Graphs are very natural forms for the presentation of the network of matter flows in ecosystems. An estimate equal to the sum of products of weights of oriented edges that constitute contour is suggested as a measure of the degree of closure in ecosystems. It is shown that this estimate can be uniformly applied to ecosystems of arbitrary size and configuration of flows. (C) 2003 COSPAR. Published by Elsevier Science Ltd. All rights reserved.

Аннотация: If soil-like substrate (SLS) is to be used in human life support systems with a high degree of mass closure, the rate of its gas exchange as a compartment for mineralization of plant biomass should be understood. The purpose of this study was to compare variations in CO2 gas exchange of vegetable plant communities grown on the soil-like substrate using a number of plant age groups, which determined the so-called conveyor interval. Two experimental plant communities were grown as plant conveyors with different conveyor intervals. The first plant community consisted of conveyors with intervals of 7 days for carrot and beet and 14 days for chufa sedge. The conveyor intervals in the second plant community were 14 days for carrot and beet and 28 days for chufa sedge. This study showed that increasing the number of age groups in the conveyor and, thus, increasing the frequency of adding plant waste to the SLS, decreased the range of variations in CO2 concentration in the “plant–soil-like substrate” system. However, the resultant CO2 gas exchange was shifted towards CO2 release to the atmosphere of the plant community with short conveyor intervals. The duration of the conveyor interval did not significantly affect productivity and mineral composition of plants grown on the SLS. © 2017

Аннотация: A number of approaches have been proposed and tested to transfer enzymatic reactions into the functional elements of microfluidic chips on the example of the bienzyme bioluminescent reaction involving NAD(P)H:FMN-oxidoreductase and luciferase. Measurement of the catalytic activity of these enzymes (under the influence of pollutants) is the basis of enzymatic bioassay of various liquids. It was found that all of the components of the reaction must be placed in the same cell of the chip to improve the reproducibility of the measurements. The use of starch gel as a carrier for immobilization and gelatin as a scaffold in the reactor of the chip enables the preservation of enzyme activity in the course of sealing the chip at room temperature. It is shown that the components of the reaction should be vigorously stirred in a microfluidic chip reactor to improve the efficiency of the analysis. As a result of the studies, a prototype of microfluidic chip based on the enzymatic bioluminescent reaction is proposed. It is characterized by a detection limit of copper sulfate of 3 mu M that corresponds to the sensitivity of traditional lux-biosensors based on living cells. The analysis time is reduced to 1 min, and the analysis can be performed by individuals without special laboratory skills.

A significant difference was observed in these groups of patterns, indicating a qualitative difference in game dynamics apparently due to the qualitatively different dynamic patterns of neuron excitations of the networks. A similar splitting of all players into two groups was found by other authors for human players, which differ in terms of the reflection availability. By this, we can assume that one of the causes of the splitting is that the presence of reflection in a particular group of recurrent neural networks dramatically changes the game meta-strategy. © Published under licence by IOP Publishing Ltd.

This fact allows us to simplify the objective of the concept of 'neural correlates of consciousness' and to focus investigations on reflection itself. Reflexive games are the concentrated and pure embodiment of reflection manifestation without the addition of other higher cognitive functions.
