Аннотация: The Yenisei River is contaminated with artificial radionuclides released by one of the Russian nuclear plants. The aquatic plants growing in the radioactively contaminated parts of the river contain artificial radionuclides. The aim of the study was to investigate accumulation of artificial radionuclides and stable elements by submerged plants of the Yenisei River and estimate the strength of their binding to plant biomass by using a new sequential extraction scheme. The aquatic plants sampled were: Potamogeton lucens, Fontinalis antipyretica, and Batrachium kauffmanii. Gamma-spectrometric analysis of the samples of aquatic plants has revealed more than 20 radionuclides. We also investigated the chemical fractionation of radionuclides and stable elements in the biomass and rated radionuclides and stable elements based on their distribution in biomass. The greatest number of radionuclides strongly bound to biomass cell structures was found for Potamogeton lucens and the smallest for Batrachium kauffmanii. For Fontinalis antipyretica, the number of distribution patterns that were similar for both radioactive isotopes and their stable counterparts was greater than for the other studied species. The transuranic elements 239Np and 241Am were found in the intracellular fraction of the biomass, and this suggested their active accumulation by the plants. В© 2011 American Chemical Society.

Аннотация: A feature of meromictic lakes is that several physicochemical and biological gradients affect the vertical distribution of different organisms. The vertical stratification of physical, chemical and biological components in saline, fishless meromictic lakes Shira and Shunet (Siberia, Russia) is quite different mainly because both mean depth and maximum depth of lakes differ as well as their salinity levels differ. The chemocline of the Lake Shira, as in many meromictic lakes, is inhabited by bacterial community consisting of purple sulphur and heterotrophic bacteria. As the depth of the chemocline is variable, the bacterial community does not attain high densities. The mixolimnion in Lake Shira, which is thermally stratified in summer, also creates different habitat for various species. The distribution of phytoplankton is non-uniform with its biomass peak in the metalimnion. The distribution of zooplankton is also heterogeneous with rotifers and juvenile copepods inhabiting the warmer epilimnion and older copepods found in the cold but oxic hypolimnion. The amphipod Gammarus lacustris which can be assigned to the higher trophic link in the fishless lake's ecosystem, such as Lake Shira, is also distributed non-uniformly, with its peak density generally observed in the thermocline region. The chemocline in Lake Shunet is located at the depth of 5 m, and unlike in Lake Shira, due to a sharp salinity gradient between the mixolimnion and monimolimnion, this depth is very stable. The mixolimnion in Lake Shunet is relatively shallow and the chemocline is inhabited by (1) an extremely dense bacterial community; (2) a population of Cryptomonas sp.; and (3) ciliate community comprising several species. As the mixolimnion of Lake Shunet is not thermally stratified for long period, the phytoplankton and zooplankton populations are not vertically stratified. The gammarids, however, tend to concentrate in a narrow layer located 1-2 m above the chemocline. We believe that in addition to vertical inhomogeneities of both physicochemical parameters, biological and physical factors also play a role in maintaining these inhomogeneities. We conclude that the stratified distributions of the major food web components will have several implications for ecosystem structure and dynamics. Trophic interactions as well as mass and energy flows can be significantly impacted by such heterogeneous distributions. Species spatially separated even by relatively short distances, say a few centimetres will not directly compete. Importantly, we demonstrate that not only bacteria, phytoflagellates and ciliate tend to concentrate in thin layers but also larger-sized species such Gammarus (amphipods) can also under certain environmental conditions have stratified distribution with maxima in relatively thin layer. As the vertical structure of the lake ecosystem is rather complex in such stratified lakes as ours, the strategy of research, including sampling techniques, should consider potentially variable and non-homogeneous distributions. Аннотация: In meromictic lakes such as Lake Shira, horizontal inhomogeneity is small in comparison with vertical gradients. To determine the vertical distribution of temperature, salinity, and density of water in a deep zone of a Lake Shira, or other saline lakes, a one-dimensional (in vertical direction) mathematical model is presented. A special feature of this model is that it takes into account the process of ice formation. The model of ice formation is based on the one-phase Stefan problem with the linear temperature distribution in the solid phase. A convective mixed layer is formed under an ice cover due to salt extraction in the ice formation process. To obtain analytical solutions for the vertical distribution of temperature, salinity, and density of water, we use a scheme of vertical structure in the form of several layers. In spring, the ice melts as top and bottom. These processes are taken into account in the model. The calculated profiles of salinity and temperature of Shira Lake are in good agreement with field measurement data for each season. Additionally, we focussed on the redox zone, which is the zone in which the aerobic layers of a water column meet the anaerobic ones. Hyperactivity of plankton communities is observed in this zone in lakes with hydrogen sulphide monimolimnion, and Lake Shira is among them. The location of the redox zone in the lake, which is estimated from field measurements, coincides with a sharp increase in density (the pycnocline) during autumn and winter. During spring and summer, the redox zone is deeper than the pycnocline. The location of pycnocline calculated with the hydro physical model is in good agreement with field measurement data. Аннотация: A one-dimensional ecological model of the meromictic brackish Lake Shira (Russia, Khakasia) was developed. The model incorporates state-of-the-art knowledge about the functioning of the lake ecosystem using the most recent field observations and ideas from PCLake, a general ecosystem model of shallow freshwater lakes. The model of Lake Shira presented here takes into account the vertical dynamics of biomasses of the main species of algae, zooplankton and microbial community, as well as the dynamics of oxygen, detritus, nutrients and hydrogen sulphide from spring to autumn. Solar radiation, temperature and diffusion are modelled using real meteorological data. The parameters of the model were calibrated to the field data, after applying different methods of sensitivity analysis to the model. The resulting patterns of phytoplankton and nutrients dynamics show a good qualitative and quantitative agreement with the field observations during the whole summer season. Results are less satisfactory with respect to the vertical distribution of zooplankton biomass. We hypothesize that this is due to the fact that the current model does not take the sex and age structure of zooplankton into account. The dynamics of oxygen, hydrogen sulphide and the modelled positions of the chemocline and thermocline are again in good agreement with field data. This resemblance confirms the validity of the approach we took in the model regarding the main physical, chemical and ecological processes. This general model opens the way for checking various hypotheses on the functioning of the Lake Shira ecosystem in future investigations and for analysing options for management of this economically important lake. Аннотация: The paper summarises the results of a three-year research study (European Union Grant: INTAS 97-0519) aimed at investigating the planktonic populations and trophic organization of the Lake Shira ecosystem - a saline lake in Khakasia, Siberia. The lake exhibits a stable summer-autumn stratification of the chemical-biological components. The mechanisms responsible for the 'blooming' of phytoplankton in the deeper layers were investigated in greater detail, using data from both field and laboratory experiments. The spectra of nutrition were examined to estimate the relationships between the specific growth rates of the hydrobionts and the influence of the limiting factors: light, nutrients. The observed heterotrophic capability of a metalimnetic phytoplankton population might help explain the development in the deeper waters of Lyngbya contorta. The scheme of trophic interactions was put up, based on the assessment of the carbon pools and carbon flows in the pelagic zone of the lake. A mathematical model of the vertical structure of the lake's plankton populations was constructed, using the ecosystem description and data of vertical turbulent diffusion. The role of light and nutrient limitations and grazing mortality in forming the vertical inhomogeneities, particularly in lowering the depth of the maximal cyanobacterial biomass, has been demonstrated. The theoretical curves for the stratification of chemical and biological parameters have been brought in conformity with the field observations, e.g. for the different patterns of the peaks, and for the biomass maxima of cyanobacteria, purple and green sulphur bacteria, oxygen, and hydrogen sulphide. The calculations revealed that for an adequate assessment of the parameters for the hydrogen sulphide zone it is necessary to introduce flows of allochthonous organic matter. Based on the form of the sulphur distribution curve, the allochthonous input of organic matter and the inflow of hydrogen sulphide from the bottom have been theoretically discriminated for the first time. It has also been ascertained that irrespective of the depth the allochthonous substances limiting bacterial growth, the bacteria are uniformly distributed over depth and can serve as an indicator of the presence of limitation (the effect of autostabilisation in space). Of indisputable interest to limnology are the specific methods developed for understanding the functioning of Lake Shira ecosystem. These include the autostabilisation of the limiting factors, the on-the-spot fluorescent method of determining the three classes of microalgae, the algal mixotrophy and the planktonic population interactions and feedbacks, and development of a more sensitive, bioluminescent method for mapping the nonhomogeneities. Owing to a balanced combination of classical approaches (field observations, in situ data on production-decomposition) and the more recent ones (satellite monitoring, biophysical methods of estimating interactions of populations, mathematical models based on the field and experimental data), many of the structural-function relationships in the ecosystem can now be explained, and the models can provide 'mutual control and mutual agreement' between the data collected using different approaches.

Аннотация: Data on the seasonal changes in vertical heterogeneity of the physical-chemical and biological parameters of the thermally stratified Shira Lake ecosystem (Khakasia, Siberia) in 1996-2000 have been analyzed. The interaction mechanisms involving: (1) The plankton populations in aerobic and anaerobic zones, involving the cycling of carbon and sulphur, (2) the primary production limitation (by light and phosphorus) and inhibition (by light), and (3) the kinetic characteristics of plankton populations have been elucidated. A mathematical model of the vertical structure of the lake's plankton populations, based on the ecosystem description and on vertical turbulent diffusion of the matter, has been constructed. The green alga Dictyosphaerium tetrachotomum (Chlorophyta) and the cyanobacterium Lyngbya contorta (Cyanophyta), which dominated the phytoplankton biomass, were taken as oxygen producers. Arctodiaptomus salinus (a calanoid copepod) has been assumed as the main grazer in Shira Lake as it dominated the zooplankton biomass. Four groups of microorganisms involved in the sulphur cycle formation have been distinguished: sulphur, sulphur purple, sulphur green and SRB. H2S is oxidized to sulphate (only the green sulphur bacteria oxidize it to sulphur), and sulphate is reduced to H2S, forming neither sulphur nor its water-soluble compounds. The role of grazing, light and nutrient limitation, in forming the vertical inhomogeneities, particularly in lowering the depth of the maximal cyanobacterial biomass, has been demonstrated. When the model takes into account both light limitation and nutrient limitation of algal growth by P and consumption of algae by crustaceans: (a) in the scenario where the P is formed only by the cycling and decomposition of autochthonous organic matter, both the green algae and cyanobacteria are eliminated; (b) in the scenario involving an additional P flux in the deep water layers the peak of the cyanobacteria is at a depth of 10 m, and its amplitude is close to the one observed in the lake. The position of the peak remains stable owing to the 'double' limitation mechanism: light 'from above' and P 'from below'. Another mechanism responsible for the deep position of the peak of cyanobacteria was analyzed mathematically based on the model involving the experimentally proven assumption of the growth inhibition by light in the epilimnion and the light limitation in the hypolimnion. The main result is: the peak is positioned stable at its depth and does not change with time. The analytical and numerical calculations made for this positioning mechanism yielded the formulae relating the depth of the maximum of algal biomass, the 'width' of the peak base and the peak amplitude and a number of parameters (algae elimination, turbulent diffusion coefficient, sedimentation rate, light extinction coefficient and light intensity). The theoretical curves for the stratification of chemical and biological parameters have been brought in conformity with field observations, e.g. for the different patterns for the peaks, and the biomass maxima of cyanobacteria, purple and green sulphur bacteria, oxygen, and hydrogen sulphide. The calculations revealed that for an adequate assessment of the parameters for the hydrogen sulphide zone it is necessary to introduce flows of allochthonous organic matter. For the first time, theoretically, based on the form of the sulphur distribution curve, the allochthonous input of organic matter and the inflow of hydrogen sulphide from the bottom have been discriminated. The theoretical limit for the depth up to which the hydrogen-sulphide zone can ascend under the impact of allochthonous organic loading, has been determined.

Аннотация: Microparticles made from degradable polyhydroxyalkanoates of different chemical compositions a homopolymer of 3-hydroxybutyric acid, copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids (P3HB/4HB), 3-hydroxybutyric and 3-hydroxyvaleric acids (P3HB/3HV), 3-hydroxybutyric and 3-hydroxyhexanoic acids (P3HB/3HHx) were prepared using the solvent evaporation technique, from double emulsions. The study addresses the influence of the chemical compositions on the size and ?-potential of microparticles. P3HB microparticles loaded with doxorubicin have been prepared and investigated. Their average diameter and ?-potential have been found to be dependent upon the level of loading (1, 5, and 10 % of the polymer mass). Investigation of the in vitro drug release behavior showed that the total drug released from the microparticle into the medium increased with mass concentration of the drug. In this study mouse fibroblast NIH 3T3 cells were cultivated on PHA microparticles, and results of using fluorescent DAPI DNA stain, and MTT assay showed that microparticles prepared from PHAs of different chemical compositions did not exhibit cytotoxicity to cells cultured on them and proved to be highly biocompatible. Cell attachment and proliferation on PHA microparticles were similar to those on polystyrene. The cytostatic drug encapsulated in P3HB/3HV microparticles has been proven to be effective against HeLa tumor cells. В© 2013 Springer Science+Business Media New York.

Аннотация: A set of polyhydroxyalkanoates are synthesized, and a comparative study of their physicochemical properties is performed. The molecular masses and polydispersities of polyhydroxyalkanoates are found to be independent of their chemical structures. It is shown that the temperature characteristics and degrees of crystallinity of polyhydroxyalkanoates are affected by the chemical compositions of the monomers and their quantitative contents in the polymers. The incorporation of 4-hydroxybutyrate, 3-hydroxyvalerate, and 3-hydroxyhexanoate units into the chain of poly(3-hydroxybutyrate) decreases its melting point and thermal degradation temperature relative to these parameters of a homogeneous poly(3-hydroxybutyrate) sample (175 В± 5 C and 275 В± 5 C, respectively). The higher the content of the second monomer units in the poly(3-hydroxybutirate) chain, the greater the changes. The degrees of crystallinity of polyhydroxyalkanoate copolymers are generally lower than that of poly(3-hydroxybutyrate) (75 В± 5%). The effect on the ratio of the amorphous and crystalline phases of the copolymer samples becomes more pronounced in the series 3-hydroxy-valerate-3-hydroxyhexanoate-4- hydroxybutyrate. Аннотация: The effect of laser irradiation on the properties of the surface of films prepared from a bioresorbable polymer poly(hydroxybuturate) has been studied. To determine the spectral region of the polymer optimal for the effective action of radiation on electron molecular bonds, theoretical investigations have been performed, which have shown that, for modifying the surface of PHB scaffolds, it is expedient to use a vacuum laser at a wavelength of 160 nm. Using laser irradiation at a power from 3 to 30 W, a series of films with modified surface, from roughnesses to perforations, have been obtained. The microstructure and properties of the film surface depending on the mode of irradiation have been examined, and conditions have been found under which the contact marginal angles of film wetting with water can be decreased to 50В° (compared with 76-80В° in starting products). Thus, conditions of laser treatment of PHB scaffolds have been theoretically substantiated and experimentally realized that provide a beneficial effect on the properties of the surface without destroying the structure of the material. Аннотация: Class I polyhydroxyalkanoic acid (PHA) synthase gene (phaC) of Ralstonia eutropha strain B5786 was cloned and characterized. R. eutropha B5786 features the ability to synthesize multicomponent PHAs with short- and medium-chain-length monomers from simple carbohydrate substrate. A correlation was made between the molecular structure of PHA synthase and substrate specificity and the ability of strain-producers to accumulate PHAs of this or that structure. A strong similarity of PHA synthase of R. eutropha strain B5786 with PHA synthase of R. eutropha strain H16, which, as opposed to strain B5786, enables to incorporate medium chain length PHAs if hexanoate is used as carbon source, exhibited 99%. A correlation between the structure of PHA synthase of B5786 strain with synthases of microorganisms which synthesize short and medium chain length PHAs similarly to B5786 strain, showed an identity level from 26 to 41% (homology with synthase of Rhodospirillum rubrum makes 41%, Ectothiorhodospira shaposhnikovii makes 26%, Aeromonas punctata makes 40%, Thiococcus pfennigii makes 28%, Rhodococcus ruber makes 38%, and with PhaCl and PhaC2 synthases of Pseudomonas sp. 61-3 makes 34 and 37%, respectively). This allows for speaking about the absence of a direct connection between the molecular organization of PHA synthases and their functional abilities, namely, the ability to synthesize PHAs of a particular composition. Аннотация: Microparticles obtained by using 14C-labeled resorbable hydroxyaminobutyric acid polymer were injected into the caudal vein of laboratory animals without negative aftereffects for their growth and development and without changes in the macro- and microstructure of organs and tissues. The distribution of microparticles in the viscera and the dynamics of accumulation of carbon-containing polymer degradation products in the viscera were studied. The main targets for the particles are liver tissues, as well as renal and splenic tissues. The polymeric matrix of the microparticles is most actively destroyed in the spleen and liver. The presence of high-molecular- weight polymeric matrix in organs indicates the integrity of microparticles and the possibility of long-term (up to 12 weeks) functioning of polymeric particles in vivo. Аннотация: The ability of marine luminescent bacteria to synthesize polyesters of hydroxycarboxylic acids (polyhydroxyalkanoates, PHA) as reserve macromolecules was studied. Twenty strains from the collection of the luminescent bacteria CCIBSO (WDCM839) of the Institute of Biophysics, Siberian Branch, Russian Academy of Sciences, assigned to different taxa (Photobacterium leiognathi, Ph. phosphoreum, Vibrio harveyi, and V. fischeri) were analyzed. The most productive strains were identified, and the conditions ensuring high polymer yields in batch culture (40-70% of the cell dry mass weight) were determined. The capacity for synthesizing two-and three-component polymers containing hydroxybutyric acid as the main monomer and hydroxyvaleric and hydroxyhexanoic acids was revealed in Ph. leiognathi and V. harveyi strains. The results allow luminescent microorganisms to be regarded as new producers of multicomponent polyhydroxyalkanoates. В© 2008 MAIK Nauka.

Аннотация: During the summers of 1999-2001 the dynamics of polyhydroxyalkanoate degradation in a small recreational eutrophic reservoir was studied experimentally. It has been shown that biodegradation of polyhydroxyalkanoates in the environment is determined by the structure and physicochemical properties of the polymer and by local weather conditions, which influence the state of the aquatic ecosystem. Species (clones) of bacteria able to utilize polyhydroxyalkanoates in the reservoir were identified using molecular phylogenetic analysis of 16S rRNA genes. В© 2007 Elsevier Ltd. Аннотация: Samples of a hybrid composite of polyhydroxybutyrate (PHB), a biodegradable polyester, and hydroxyapatite (HA), with different PHB/HA ratios, have been prepared using mechanical-physical method. Electron microscopy, X-ray structure analysis and differential thermal analysis have been used to investigate the structure and physicochemical properties of the composite, depending on the PHB/HA ratio. The properties of the surface of the HA-loaded composite are significantly different from those of the pure polymer. As the HA percentage in the composite increases, free interface energy, the cohesive force, i.e., the strength of the adhesive bond between the composite surface and the water phase, and surface wettability increase. The HA percentage of the composite does not influence its melting temperature, but affects the temperature for the onset of decomposition: as the HA content increases from 0 to 10% (w/w), Td decreases from 260В°C to 225В°C. The degree of crystallinity of PHB/HA increases from 77% to 89% with an increase in the HA fraction from 10% to 50%. Functional properties of the composites have been investigated in vitro and in vivo. The best parameters of growth and differentiation of murine marrow osteoblasts are registered on PHB/HA samples containing 10% and 20% HA. In ectopic bone formation assay it has been proven that the hybrid PHB/HA composites can function as scaffolds and that bone tissue develops on their surface and in pores. В© VSP 2006.

Аннотация: The biodegradability of oriented fibers made of polyhydroxybutyrate (P(3HB)) and its co-polymer with ?-hydroxyvalerate (P(3HB-co-3HV)) was investigated in buffer solutions and in biological media in vitro and in vivo. The fibers of both polymer types demonstrated resistance to hydrolytic degradation in buffer solutions at 38В°C and pH from 4.5 to 7.0 (for up to 180 days). It has been found that the biodegradation of the fibers in vitro in blood and serum and in vivo is accompanied by weight losses and minor changes in the microstructure with no significant losses in the tensile strength over a long time (up to 180 days). The biodegradation rate of the less crystalline co-polymer P(3HB-co-3HV) fibers was 1.4-2.0-times higher than that of the homopolymer P(3HB). It has also been shown that the degradation of the fibers in vivo is influenced both by tissue fluid enzymes and cells (macrophages and foreign-body giant cells). The fibers were eroded on the surface only with no gross defects and no dramatic effects on their mechanical performance. В© VSP 2005.

Аннотация: A series of two-component polyhydroxyalkanoates composed of hydroxybutyrate-hydroxyvalerate copolymers with different monomer ratio was obtained with the use of bacteria Ralstonia eutropha B5786. The properties of the polyhydroxyalkanoates in comparison with the homopolymer of hydroxybutyric acid were studied by X-ray diffraction analysis, IR spectroscopy, differential thermal analysis, and viscometry. The ratio of crystalline to amorphous phase in the copolymers tends to unity with increasing hydroxyvalerate content. This is accompanied by a decrease in the degree of crystallinity of the copolymers from 70-80 to 45-50%, the dependence is virtually linear within the range, of hydroxyvalerate mole fraction from several to 25-30 mol%. Thermal characteristics, melting temperature (Tm) and decomposition temperature (Td), of the polyhydroxyalkanoate copolymers are lower than those for polyhydroxybutyrate, whose Tm and Td are 168-170 and 260-265В°C, respectively. Both parameters decrease to 150-160 and 200-220В°C, respectively, when the hydroxyvalerate mole fraction is raised. Аннотация: Polyhydroxyalkanoate (PHA) sutures were implanted to test animals intramuscularly, and tissue reaction was investigated and compared with the reaction to silk and catgut. Tested monofilament sutures made of PHAs of two types polyhydroxybutyrate (PHB) and a copolymer of hydroxybutyrate and hydroxyvalerate (PHV) featured the strength necessary for the healing of muscle-fascial wounds. The reaction of tissues to polymeric implants was similar to their reaction to silk and was less pronounced than the reaction to catgut; it was expressed in a transient post-traumatic inflammation (up to four weeks) and the formation of a fibrous capsule less than 200 ?m thick, which became as thin as 4060 ?m after 16 weeks, in the course of reverse development. Macrophages and foreign-body giant cells with a high activity of acid phosphatase were actively involved in this process. PHB and PHB/PHV sutures implanted intramuscularly for an extended period (up to one year) did not cause any acute vascular reaction at the site of implantation or any adverse events, such as suppurative inflammation, necrosis, calcification of the fibrous capsule or malignant tumor formation. No statistically significant differences were revealed in the tissue response to polymer sutures of the two types. Capsules around silk and catgut sutures did not become significantly thinner.

Аннотация: The structure of liquid hydrocarbons and fatty acids produced by the green alga Botryococcus was identified. Two representatives of this alga, Botryococcus braunii Kutz, strain IPPAS H-252, introduced into culture earlier and an organism isolated for the first time from the Shira Lake, were used for this identification. Fatty acid composition of B. braunii, strain H-252, lipids was characterized by a high content of trienoic acids of C16-C18 series. The hydrocarbon composition of this strain was represented by straight-chain and branched-chain C14-C28 components; long-chain linear aliphatic C20-C27 hydrocarbons (54.4%) and 2,6,10,14-tetramethylhexadecane (20.5%) predominated among them. The strain H-252 differed in its fatty acid and hydrocarbon composition from the strains described earlier as Botryococcus braunii. The fatty acid composition of the Botryococcus isolate was represented mainly by C12-C32 saturated and monoenoic acids. The hydrocarbons formed by this isolate were represented by dienoic and trienoic components. C29 (48.9-56.3%) and C31 (11.1-16.3%) hydrocarbons predominated among the C23-C31 dienoic hydrocarbons, and C27, C29, and C31 trienoic hydrocarbons comprised 2.5-2.6% of total hydrocarbons. This type of hydrocarbons and the lipid fatty acid composition were characteristic for the race A of B. braunii.
