Аннотация: The study was done to determine concentrations of radionuclides and heavy metals and to evaluate the frequency of chromosomal aberrations in samples of Elodea canadensis, a submerged plant, collected in different parts of the Yenisei River. Samples were collected in the area subjected to radioactive impact of theMining-and-Chemical Combine (MCC) at Zheleznogorsk and in the control areas, upstream of the MCC. The investigations showed that Elodea biomass in the area affected by MCC operation contained a long inventory of artificial radionuclides typical of the MCC discharges. Upstream of the MCC, in the control sampling areas, the sediments and the Elodea biomass contained only one artificial radionuclide - 137Cs. Thus, the exposure doses to Elodea shoots and roots upstream of the MCC are small (not more than 8 ?Gy/d) and the main contribution to them is made by natural radionuclides. At the MCC discharge site (the village of Atamanovo) and downstream of it, the total dose rate increases almost an order of magnitude, reaching its maximal values - 72 ?Gy/d for Elodea shoots and 58 ? Gy/d for roots. Cytogenetic investigations of Elodea roots showed that at the MCC discharge site (the village of Atamanovo) and downstream of it the occurrence of chromosomal aberrations in ana-telophase and metaphase cells of Elodea was considerably higher than in the control area. It is highly probable that this simultaneous dramatic increase in the total exposure rate and the occurrence of chromosomal aberrations in Elodea is associated with the radiation factor. It is suggested that Elodea is affected not only by the radiation factor but also by the chemical factor - toxicity of heavy metals. В© 2009 EDP Sciences.

Аннотация: A set of bioluminescent tests was developed to monitor water quality in natural and laboratory ecosystems. It consisted of four bioluminescent systems: luminous bacteria, coupled enzyme system NADH:FMN-oxidoreductase-luciferase and triplet enzyme systems with alcohol dehydrogenase and trypsin. The set of biotests was applied for a small forest pond (Siberia, Russia), laboratory microecosystems polluted with benzoquinone and a batch culture of blue-green algae. Thereby effects of natural water compared to those of models of heavy pollution and "bloom" of blue-greens on the bioluminescent tests were revealed. The set of biotests was not affected by a natural seasonal variability of water quality in the unpolluted pond, but responded to the heavy pollution and the "bloom" of blue-greens. The set of biotests could be recommended as the alarm test to control the acute toxicity of natural water bodies. В© 2001 Elsevier Science Ltd.

Аннотация: The life support systems (LSS) for long-term missions are to use cycling-recycling systems, including biological recycling. Higher plants are the traditional regenerator of air and producer of food. They should be used in many successive generations of their reproduction in LSS. Studies of influence of microbial associations on germination of wheat seeds and on growth of seedlings under impact of heavy metals are necessary because of migration of heavy metals in LSS. Microbial associations are able to stimulate growth of plants, to protect them from pathogenic organisms and from toxicity of heavy metal salts. The goal of this work was to investigate effect of microbial associations on the germination of wheat seeds and on the growth of seedlings under impact of different concentrations of ZnSO4. The results of investigations showed that:(1)Zinc salt had an adverse effect on germination of wheat seeds, beginning with concentrations of 8 MPC (Maximum Permissible Concentration) and higher.(2)Microbial associations (concentrations -104 to 107 cells/ml) were able to decrease (partly or completely) the adverse effect of ZnSO4 on germination of wheat seeds.(3)Concentrations (104-107 cells/ml) of microbial associations were able to decrease partly the adverse effect of zinc salts (intervals: from 1 to 32 MPS{cyrillic}) on the growth and development of wheat plantlets during heterotrophic phase.(4)The root system of plants was more sensitive to the adverse effect of ZnSO4 than shoots of plants. Аннотация: Waters of the Yenisei River, certain rivers and lakes of the Altai Territory, and effluents of some industrial factories in Krasnoyarsk were studied by luminous bacteria biotest Microbiosensor B17 677F. The lyophilized luminous bacteria Photobacterium phosphoreum from the IBSO collection were used to design this biotest. The bioluminescent test is based on bioluminescence quenching resulting from the action of water samples on luminous bacteria. The test results indicated locations and zones of impaired water quality. The heaviest pollution of water in the Yenisei River was recorded in the zones 0-116 km downstream from Krasnoyarsk (Krasnoyarsk and satellite towns). The effluents of most factories were found to be toxic. Underground and surface waters of some areas of the Altai Territory had different toxicity levels; there were deviations from the norm in most water samples taken from the different lakes and rivers. The data from this study show that the luminous bacteria biotest is simple and convenient, and that the results obtained are within acceptable levels of accuracy for the evaluation of the toxicity of effluent and river water. It takes no more than 30 min to do the biotest. It can be used in ecological monitoring like the Microtox toxicity test. В© 1998 John Wiley & Sons, Inc.

Аннотация: The bacterial bioluminescence has high sensitivity to the action of various inhibitors of biological activity. The lyophilized luminous bacteria Photobacterium phosphoreum (Microbiosensor B17 677F) and luminous strain Escherichia coli (Microbiosensor EC) from the Culture Collection IBSO were used to create bioluminescent biotests. They have been applied in ecological monitoring to determine the overall toxicity of the Yenisei and Angara Rivers and some water sources of Altai Territory. As a rule the heaviest pollution of water in studied rivers was registered near cities and settlements. The luminous bacteria biotests are simple and convenient in work, standardized and quantitative, have rapid response to actions of different substances and high sensitivity to environmental pollutants. It takes less than 30 min to do the biotest (the other biotests take 48-96 h). Copyright В© 1999 John Wiley & Sons, Ltd.

Аннотация: The inhibition of bacterial luminescence has been used in testing industrial enterprises sewage. The toxicity of the sewage is less than the total toxicity of separate components due to neutralization of quinone products of polyphenol oxidation in the reactions with the other phenol components of sewage. Toxicity increase is due to their influence on the cell membrane. Studies of cell ultrastructure confirm this fact. The studied mechanism of the complex effect allowed a more accurate forecast of the ecological situation during the discharge of phenol compounds and metals. It also showed the necessity of taking into account the complex effect of sewage components on contaminant discharge into water reservoirs.

Аннотация: The physical-chemical regularities of aromatic compounds' effects in luciferase to toxicity biotesting have been studied, The structures and physical-chemical characteristics of the toxicants and of the bioluminescent emitter were taken into account. The inhibition constants of bioluminescence intensity (I) were calculated and interpreted from the viewpoint of the energy (electron) transfer processes. The induction period (P) and the increase of the rime of the maximum light intensity (t(M)) which take place in the quinones presence, have been shown to deal with hydrogen transfer processes. The values of I, P and t(M) have been shown to be connected with a size of the quinones' aromatic and aliphatic parts, P- and t(M)-dependencies on quinone's redox potential have been demonstrated.

Аннотация: This study addresses the effects of low-level alpha-radiation on bioluminescent assay systems of different levels of organization: in vivo and in vitro. Three bioluminescent assay systems are used: intact bacteria, lyophilized bacteria, and bioluminescent system of coupled enzyme reactions. Solutions of Am-241(NO3)(3) are used as a source of alpha-radiation. It has been shown that activation processes predominate in all the three bioluminescent assay systems subjected to short-term exposure (20-55 h) and inhibition processes in the systems subjected to longer-term exposure to radiation. It has been found that these effects are caused by the radiation component of Am-241(3+) impact. The intensity of the Am-241(3+) effect on the bioluminescent assay systems has been shown to depend on the Am-241(3+) concentration, level of organization and integrity of the bioluminescent assay system. The bioluminescent assay systems in vivo have been found to be highly sensitive to Am-241(3+) (up to 10(-17) M).

Аннотация: The purpose of this study was to develop micro and nano sized drug carriers from poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV), and study the cell and skin penetration of these particles. PHBV micro/nanospheres were prepared by o/w emulsion method and were stained with a fluorescent dye, Nile Red. The particles were fractionated by centrifugation to produce different sized populations. Topography was studied by SEM and average particle size and its distribution were determined with particle sizer. Cell viability assay (MTT) was carried out using L929 fibroblastic cell line, and particle penetration into the cells were studied. Transdermal permeation of PHBV micro/nanospheres and tissue reaction were studied using a BALB/c mouse model. Skin response was evaluated histologically and amount of PHBV in skin was determined by gas chromatography-mass spectrometry. The average diameters of the PHBV micro/nanosphere batches were found to be 1.9 mu m, 426 and 166 nm. Polydispersity indices showed that the size distribution of micro sized particles was broader than the smaller ones. In vitro studies showed that the cells had a normal growth trend. MTT showed no signs of particle toxicity. The 426 and 166 nm sized PHBV spheres were seen to penetrate the cell membrane. The histological sections revealed no adverse effects. In view of this data nano and micro sized PHBV particles appeared to have potential to serve as topical and transdermal drug delivery carriers for use on aged or damaged skin or in cases of skin diseases such as psoriasis, and may even be used in gene transfer to cells.

Аннотация: Effects of Americium-241 (241Am), alpha-emitting radionuclide of high specific radioactivity, and tritium (3H), beta-emitting radionuclide, on luminous bacteria Photobacterium phosphoreum were compared. Bioluminescence intensity served as a marker of bacterial physiological activity. Three successive stages in the bioluminescence response to 241Am and 3H were found under conditions of lowdose irradiation: (1) absence of effects, (2) activation, and (3) inhibition. They were interpreted in terms of bacterial response to stressfactor as stress recognition, adaptive response/syndrome, and suppression of physiological function (i.e. radiation toxicity). Times of bioluminescence activation (TBA) and inhibition (TBI) were suggested as parameters to characterize hormesis and toxic stages in a course of chronic low-dose irradiation of the microorganisms. Values of TBA and TBI of 241Am were shorter than those of 3H, revealing higher impact of alpha-irradiation (as compared to beta-irradiation) under comparable radiation doses. Increases of peroxide concentration and NADH oxidation rates in 241Am aquatic solutions were demonstrated; these were not found in tritiated water. The results reveal a biological role of reactive oxygen species generated in water solutions as secondary products of the radioactive decay. The study provides a scientific basis for elaboration of bioluminescence-based assay to monitor radiotoxicity of alpha- and beta-emitting radionuclides in aquatic solutions. © 2014 Versita Warsaw and Springer-Verlag Wien.

Аннотация: The laboratory testing of bottom sediments (BSs) from the Yenisei River containing different concentrations of technogenic radionuclides, heavy metals, and biogenic elements (N and P) based on aquatic such plants as Elodea canadensis (Canadian waterweed) and Myriophyllum spicatum (Eurasian watermilfoil) has revealed a higher sensitivity of roots to the general quality of BSs than shoots: shoot length (9%) < root length (11%) < root number (15%) in M. spicatum; shoot length (22%) < root length (42%) < root number (44%) in E. canadensis. In contrast to M. spicatum, the growth parameters of roots and shoots in E. canadensis have differed in a significant statistical manner between most BS samples. A reverse correlation has been found between the increase in shoot length and the activity of technogenic radionuclides in BSs, which is mostly significant in E. canadensis (r (2) = 0.90-0.95, p = 0.05). Since the growth of shoots and roots in E. canadensis has turned out to be more sensitive to changes in the quality of BSs than that in M. spicatum, E. canadensis can be considered more prospective for biotesting BSs.

Аннотация: Chromosome abnormalities in ana-telophase cells of apical root meristem of aquatic plant Elodea. canadensis (elodea), sampled in 2011-2012 in the Yenisei River at a site with background level of contamination and at several sites on the stretch contaminated with artificial radionuclides, and with chemical pollutants from municipal and industrial discharges of the Krasnoyarsk city. Lowest rate (5.2%) of cells with chromosome abnormalities was registered at sampling site with background level of contamination upstream of the Krasnoyarsk, highest rate of cells with abnormalities (39.7%)-in roots of elodea sampled in bottom sediments with highest concentration of Cs-137. Sum of rates of cells with abnormalities and rates of cells with all types of abnormalities positively correlated with total concentration of artificial and natural radionuclides, with concentration of artificial radionuclides and Cs-137 in bottom sediments of the Yenisei River (r (2) = 0.91-0.96, p < 0.0005 for sum of rates of cells with abnormalities; r (2) = 0.58-0.92, p < 0.05 for all types of abnormalities).

Аннотация: Various chronic inflammatory and necrotic processes in the liver parenchyma are accompanied by pathological morphofunctional changes, which are associated with hepatocyte death and hyperplasia of the connective tissue. Regeneration of the liver parenchyma should include not only prevention of fibrosis, but also stimulation of hepatocyte prolifearation. The adrenoceptor agonist dobutamine stimulated proliferative activity of cultured hepatocytes and prevented the development of postintoxication liver cirrhosis in mice produced by chronic poisoning with CCl4.
