Аннотация: A submerged macrophyte of the Yenisei River, Elodea canadensis, was used to study the microdistribution of the artificial radionuclide 241Am among different components of the plant. The total amount of 241Am added to the experimental system was 1850 В± 31 Bq/L. The total amount of 241Am accumulated by the plants was 182 Bq per sample, or 758,333 В± 385 Bq/kg dry mass. It has been found that the major portion of 241Am accumulated by E. canadensis, up to 85%, was bound to solid components of the cells. It is observed that the microdistribution of 241Am within different components of the submerged plant E. canadensis was not uniform. 241Am distribution vary depending on the age of the leaf blades, the state of the cells and morphological features of the plant stem. Аннотация: Changes in the intracellular concentration of free ionized calcium ([Ca2+]i) control a host of cellular processes as varied as vision, muscle contraction, neuronal signal transmission, proliferation, apoptosis etc. The disturbance in Ca2+-signaling causes many severe diseases. To understand the mechanisms underlying the control by calcium and how disorder of this regulation relates to pathological conditions, it is necessary to measure [Ca2+]i. The Ca2+-regulated photoproteins which are responsible for bioluminescence of marine coelenterates have been successfully used for this purpose over the years. Here we report the results on comparative characterization of bioluminescence properties of aequorin from Aequorea Victoria, obelin from Obelia longissima, and clytin from Clytia gregaria charged by native coelenterazine and coelenterazine analogues f, i, and hcp. The comparison of specific bioluminescence activity, stability, emission spectra, stopped-flow kinetics, sensitivity to calcium, and effect of physiological concentrations of Mg2+ establishes obelin-hcp as an excellent semisynthetic photoprotein to keep track of fast intracellular Ca2+ transients. The rate of rise of its light signal on a sudden change of [Ca2+] is almost 3- and 11-fold higher than those of obelin and aequorin with native coelenterazine, respectively, and 20 times higher than that of the corresponding aequorin-hcp. In addition, obelin-hcp preserves a high specific bioluminescence activity and displays higher Ca2+-sensitivity as compared to obelin charged by native coelenterazine and sensitivity to Ca2+ comparable with those of aequorin-f and aequorin-hcp. © The Royal Society of Chemistry and Owner Societies 2015.

Аннотация: We describe a low-cost three-dimensional underwater particle tracking velocimetry system to directly measure particle settling rate and flux in low-turbulence aquatic environments. The system consists of two waterproof cameras that acquire stereoscopic videos of sinking particles at 48 frames s?1 over a tunable sampling volume of about 45 ? 25 ? 24 cm. A dedicated software package has been developed to allow evaluation of particle velocities, concentration and flux, but also of morphometric parameters such as particle area, sinking angle, shape irregularity, and density. Our method offers several advantages over traditional approaches, like sediment trap or expensive in situ camera systems: (1) it does not require beforehand particle collection and handling; (2) it is not subjected to sediment trap biases from turbulence, horizontal advection, or presence of swimmers, that may alter particulate load and flux; (3) the camera system enables faster data processing and flux computation at higher spatial resolution; (4) apart from the particle settling rates, the particle size distribution, and morphology is determined. We tested the camera system in Lake Stechlin (Germany) in low turbulence and mean flow, and analyzed the morphological properties and settling rates of particles to determine their sinking behavior. The particle flux assessed from conventional sediment trap measurements agreed well with that determined by our system. By this, the low-cost approach demonstrated its reliability in low turbulence environments and a strong potential to provide new insights into particulate carbon transport in aquatic systems. Extension of the method to more turbulent and advective conditions is also discussed. © 2019 The Authors.
