Аннотация: Dihydroneopterin aldolase (DHNA) catalyses a retroaldol reaction yielding 6-hydroxymethyl-7,8-dihydropterin, a biosynthetic precursor of the vitamin, tetrahydrofolate. The enzyme is a potential target for antimicrobial and anti-parasite chemotherapy. A gene specifying a dihydroneopterin aldolase from Arabidopsis thaliana was expressed in a recombinant Escherichia coli strain. The recombinant protein was purified to apparent homogeneity and crystallised using polyethylenglycol as the precipitating agent. The crystal structure was solved by X-ray diffraction analysis at 2.2 Angstrom resolution. The enzyme forms a D-4-symmetric homo-octamer. Each polypeptide chain is folded into a single domain comprising an antiparallel four-stranded beta-sheet and two long alpha-helices. Four monomers are arranged in a tetrameric ring, and two of these rings form a hollow cylinder. Well defined purine derivatives are found at all eight topologically equivalent active sites. The subunit fold of the enzyme is related to substructures of dihydroneopterin triphosphate epimerase, GTP cyclohydrolase I, and pyruvoyltetrahydropterin synthase, which are all involved in the biosynthesis of pteridine type cofactors, and to urate oxidase, although some members of that superfamily have no detectable sequence similarity Due to structural and mechanistical differences of DHNA in comparison with class I and class II aldolases, a new aldolase class is proposed. Аннотация: Light intensity and nitrogen concentration of nutrient solution are considered crucial for the contents of vitamin C and nitrate in cultivated leaf vegetables. We here investigated the effects of various combinations of light intensity (60,140 and 220 mu mol m(-2) s(-1)) and nitrogen concentration (7,15 and 23 mmol L-1) of the solution on the growth, photosynthetic characteristics, vitamin C and nitrate content of lettuce. Our results demonstrate that the lettuce had the largest dry biomass at the high illumination of 220 mu mol m(-2) s(-1) and low nitrogen of 7 mmol L-1. The higher light and low nitrogen also contributed to the accumulation of vitamin C and decrease of nitrate in lettuce leaves. The effect of nitrogen supply on chlorophyll concentrations was more efficient under low light than high illumination. Our results reveal that there was an obvious interaction between light intensity and nitrogen available for the photosynthesis, yield and quality of lettuce. This study provides valuable insights into the combinational regulation of light intensity and nitrogen supply for improving growth and nutritional quality of vegetables grown in greenhouse and plant factory. Аннотация: In view of previous studies, a list of 46 foods designated for use in bioregenerative life support system was composed. With the help of a computer program, daily sets of foods of plant and animal origin were compiled from the list of foods. The objective function of modeling was intended to minimize the discrepancy between the calculated values of nutrients in daily food sets and NASA nutrition requirements for long-duration missions. The independent variables in the model were the masses of foods restricted by the lower and upper bounds. It was established that a food set is able to comprise 10-46 foods with violation of the NASA nutrition requirements for iron, vitamin B5 and vitamin D daily intakes. Inclusion of 9 foods in a set resulted in a further violation of the NASA standards concerning saturated fat. As the number of foods in a set has increased from 10 to 22, the objective function decreased from 1.0736 to 1.0332, followed by a gradual increase to 1.1233, when the maximum number of foods was selected from the list of foods. The source of uncertainty in the interpretation of modeling results are the standard NASA intakes of magnesium, potassium, zinc, manganese, vitamin C, thiamin, riboflavin, niacin, vitamin B6, folate, vitamin B12, vitamin E, vitamin K and n-6 fatty acids, given as exact values. Varying the nutrient content of food sets did not significantly affect the value of the objective function. However, some solutions were infeasible, due to the violation of the NASA standard concerning saturated fat. Also, there were food sets in which the scores of sulfur-containing amino acids and threonine were below 100. In order to reliably maintain the scores of essential amino acids above 100 in a food set, it is necessary to maintain a mass ratio of “animal protein/total protein” equal to 2/3 in accordance with the requirement of NASA. © 2019 IAA

Аннотация: The purpose of the present study was to investigate the efficacy of the experimental formulations of the metribuzin (MET) and tribenuron-methyl (TBM) herbicides embedded in the matrix of degradable poly-3-hydroxybutyrate blended with wood flour in field-grown tomato and beet crops infested by weeds. There is a necessity to develop environmentally friendly and effective means to protect plants because of the shortcomings of the free herbicide forms such as the environmentally unsafe spray application of solutions and suspensions of the widespread metribuzin and tribenuron-methyl herbicides, removal from soil during watering events and rains, and transport to natural aquatic environments, where the herbicides accumulate in the trophic chains of biota. Free TBM is also rapidly inactivated in soil and metabolized to nontoxic products in plants. The efficacy of experimental formulations of metribuzin and tribenuron-methyl embedded in the matrix of degradable poly-3-hydroxybutyrate blended with wood flour was tested in field-grown tomato and beet crops infested with weeds. Application of metribuzin resulted in the highest productivity of tomatoes (2.3 kg/m(2)) and table beet (3.4 kg/m(2)), improved biometric parameters of tomato fruits and beet roots, and caused reduction in nitrate nitrogen concentrations in them. The mode of herbicide delivery did not affect sugar contents, but application of both metribuzin and tribenuron-methyl induced a 1.7-fold and 1.4-fold, respectively, increase in vitamin C concentrations in tomato fruits and beet roots relative to the vegetables grown on the subplots treated with free herbicides and the intact plants. Embedded herbicides can be used as preemergence herbicides in the field.

Аннотация: The capability of "dish" and "ingredient in dish" modeling to reduce the number of nutrition imbalances in bioregenerative life support diet was compared. Masses of dishes were assumed to be the independent variables in the 'dish' model, while in the 'ingredient in dish' model the independent variables were the total masses of the ingredients in a one-day menu and masses of ingredients in the dishes. The objective function in both models was minimization of discrepancy between the calculated nutrition intakes and the daily nutrition requirements of NASA for long duration space missions. Comparing of two models was carried out for the case of a one-day diet containing 12 dishes and 32 ingredients. It was established that the diet simulation by 'dish' model brings 6 nutrition imbalances. The use of the 'ingredient in dish' modeling has helped to reduce the number of nutrition imbalances to 3, namely, an excess of iron, vitamin A and saturated fat. Obstacles to the fulfillment of all nutrition requirements were the nomenclature and masses of ingredients in the dish recipes.

Аннотация: Streptavidin is a homotetrameric protein produced by Streptomyces avidinii, each subunit of which binds biotin (vitamin H), forming a stable complex (Kd = 10-15 M). Streptavidin-biotin coreaction is widely used in analytical systems, for targeted delivery of compounds, for affinity purification, etc. The aim of this study was to develop a rational technique to produce functionally active recombinant streptavidin. Recombinant Escherichia coli strains producing minimal core and full-sized streptavidin variants were obtained. The E. coli BL21 Codon Plus (DE3) RIPL, as host cells, and the pET19b plasmid carrying gene of minimally-sized core (miniSAV) or full-sized (SAV) streptavidin were used. Synthesis of miniSAV results in its localization as insoluble inclusion bodies. Denatured miniSAV yield was 130 mg per liter of E. coli c ulture. T he r enaturation g ives o nly 10- 15 % of the functionally active protein. Full-sized streptavidin localizes in the cytoplasm in a soluble state, but its toxicity causes low yield of the protein (10-13 mg per liter of the culture). The induction of SAV synthesis at the end of the logarithmic stage of cell growth was found to increase the yield of SAV approximately 2-fold. The yield of functionally active protein was 30 mg per liter culture. SAV was produced practically in individual state after affine chromatography on 2-iminobiotin agarose. One molecule of full-sized streptavidin bound 3.9 biotin molecules as was shown by colorimetric analysis using HABA (4-hydroxyazobenzene-2-carboxylic acid). Both streptavidins form sandwichtype complexes with biotinylated molecules in solid-phase microassay conditions. E. coli BL21 Codon Plus (DE3) RIPL/pET19bSAV strain was stable during storage with 20 % glycerol at -70 °C, which was shown by repeated two-year reseeding. The streptavidin producing strain (E. coli BL21 Codon Plus (DE3) RIPL/pET19bSAV) is deposited in the Collection for extremophile microorganisms and type cultures (Institute of Chemical Biology and Fundamental Medicine SB RAS, Novosibirsk), No. 3505. The method for producing functionally active recombinant streptavidin developed in this study ensures its availability for biotechnological research. © Siberian Federal University.
