Аннотация: The integrated study addressing biodegradation of microbial linear polyesters of hydroxyalkanoic acids (polyhydroxyalkanoates, PHAs) in tropical conditions by microbial communities of Vietnamese soils was performed in locations close to Hanoi and Nha Trang, which differed in their weather conditions and microbial communities. It shows that PHA degradation in tropical soils is influenced by polymer chemical composition, specimen shape, and microbial characteristics. The homopolymer of 3-hydroxybutyric acid is degraded at higher rates than the copolymer of 3-hydroxybutyric and 3-hydroxyvaleric acids. The average rates of mass loss were 0.04-0.33% per day for films and 0.02-0.18% for compact pellets. PHA degradation was accompanied by a decrease in the polymer molecular mass and, usually, an increase in the degree of crystallinity, suggesting preferential degradation of the amorphous phase. Under the study conditions, representatives of the bacterial genera Burkholderia, Bacillus, Cupriavidus, Mycobacterium, and Nocardiopsis and such micromycetes as Acremonium, Gongronella, Paecilomyces, and Penicillium, Trichoderma have been identified as major PHA degraders. Аннотация: The biodegradable polymer poly(3-hydroxybutyrate-co-3-hydroxyvalerate) has been used to design experimental sustained-release formulations of the herbicide Zellek Super in the form of films and microgranules. The kinetics of polymer degradation and the dynamics of herbicide release show that the rate and extent of herbicide release from the polymer matrix into the soil depends on the geometry of the carrier and the proportion of the pesticide loaded into it (polymer/pesticide mass ratio). Experiments with the creeping bentgrass (Agrostis stolonifera L.) show that the formulations of the herbicide Zellek Super constructed as microgranules and films can be successfully used to suppress the growth of grasses. This study is the first to demonstrate that biodegradable polyhydroxyalkanoates can be used effectively to construct environmentally friendly sustained-release PHA-herbicide systems that can be placed into the soil together with seeds.

Аннотация: Biodegradation of microbial linear polymers of hydroxyalkanoic acids (polyhydroxyalkanoates, PHAs) by soil microbial communities of different structures has been studied during two field seasons in different weather conditions. This process was shown to be influenced by the polymer chemical composition, temperature, humidity, and the microbial soil component. The PHA degradation was accompanied by a decrease in the polymer molecular weight and an increase in the degree of crystallinity, indicating the preferential destruction of the amorphous phase compared to the crystalline one. The quantity of the true PHA destructors developing at the surface of the polymer samples was lower than the quantity of accompanying bacteria. The dominant PHA degrading microorganisms under the test conditions were identified as bacteria of the genera Variovorax, Stenotrophomonas, Acinetobacter, Pseudomonas, Bacillus, and Xanthomonas and as micromycetes from Penicillium, Paecilomyces, Acremonium, Verticillium, and Zygosporium.

Аннотация: The effect of different cultivation parameters on the specific activities of soluble and membrane-bound hydrogenases was studied in Alcaligenes eutrophus Z-1 cells. These activities were shown to be dependent primarily on the availability of electrons from the substrate used. Changes in cultivation temperature and oxygen concentration versus the optimal values as well as addition of low amounts of carbon monoxide to the gas phase exerted a lesser effect on the specific hydrogenase activities. Autotrophic and heterotrophic methods were developed for growing A. eutrophus Z-1 cells with a high hydrogenase activity.

The last factor is critical in the maintenance of recombinant plasmids in transgenic microorganisms.

Аннотация: In experimental aquatic microcosms (AMCs), the population of the Escherichia coli strain Z905 harboring the recombinant plasmid pPHL7 (Ap(r)Lux(+)) was found to gradually accumulate AMC-adapted cells, which retained the plasmid but differed from the original cells in some biochemical and physiological characteristics. Both the original and the AMC-adapted E. coil cells could coexist with the native AMC microflora for one year or longer. When introduced into AMCs together with native pseudomonads, the AMC-adapted E. coil Z905-33 (pPHL7) cells were more competitive than the nonadapted cells.

Аннотация: Modeling of transgenic microorganism introduction into small man-made ecosystems can help forecast changes in expression of cloned genes under different conditions of existence. Introduction of the E. coli Z905/pPHL7 strain containing a plasmid with luminescent system genes of luminous bacteria led to changes in cell and colony morphology, reduction in metabolic activity of cells, and, as a result, a lower level of expression of cloned gene. A low concentration of nutrients has been shown to favor greatly the phenotypic change of cells of the recombinant strain. Expression of cloned genes changed due to: a lower concentration of plasmid DNA, a change in regulation of cloned genes, and a change in cells of biosynthesis of substrates needed for expression of luminescent genes. The conducted investigations can provide a basis for the use of marker transgenic microorganisms in closed ecosystems of different types. (C) 2001 COSPAR. Published by Elsevier Science Ltd. Аннотация: Microbiological and biogeochemical studies of the meromictic saline Lake Shira (Khakasia) were conducted. In the upper part of the hydrogen-sulfide zone, at a depth of 13.5-14 m, there was a pale pink layer of water due to the development of purple bacteria (6 x 10(5) cells/ml), which were assigned by their morphological and spectral characteristics to Lamprocystis purpurea (formerly Amoebobacter purpureus). In August, the production of organic matter (OM) in Lake Shira was estimated to be 943 mg C/(m(2) day). The contribution of anoxygenic photosynthesis was insignificant (about 7% of the total OM production). The share of bacterial chemosynthesis was still less (no more than 2%). In the anaerobic zone, the community of sulfate-reducing bacteria played a decisive role in the terminal decomposition of OM. The maximal rates of sulfate reduction were observed in the near-bottom water (114 mug S/(1 day)) and in the surface layer of bottom sediments (901 mug S/(dm(3) day)). The daily expenditure of C-org for sulfate reduction was 73% of C-org formed daily in the processes of oxygenic and anoxygenic photosynthesis and bacterial chemosynthesis. The profile of methane distribution in the water column and bottom sediments was typical of meromictic reservoirs. The methane content in the water column increased beginning with the thermocline (7-8 m) and reached maximum values in the near-bottom water (17 mul/l). In bottom sediments, the greatest methane concentrations (57 mul/l) were observed in the surface layer (0-3 cm). The integral rate of methane formation in the water column and bottom sediments was almost an order of magnitude higher than the rate of its oxidation by aerobic and anaerobic methanotrophic microorganisms.

Аннотация: The composition and content of fatty acids (FAs) in total lipids, triacylglycerols (TAG) and polar lipids (PL) in dominant groups of benthic invertebrates: gammarids (Gammaridae, Amphipoda), chironomid larvae (Chironomidae, Diptera), caddisfly larvae (Trichoptera) and mayfly larvae (Ephemeroptera) were studied in the Yenisei river. For the first time data on the FA composition of species belonging to Trichoptera (Insecta) are presented. The groups of aquatic insect larvae and gammarids weakly differed in total content of essential polyunsaturated fatty acids (PUFAs). Hence, the strong invasion of gammarids which occurred in the last decades in the Yenisei river should not result in a decrease in potential yield of essential PUFA in the ecosystem and corresponding decrease in food resource quality for fish in respect to PUFA content. Significant differences in biomarker FAs in TAG were found which correlated to specific food sources. Different levels of long-chain PUFA in PL of the invertebrates are discussed in relation to the genetic ability of particular taxa to form these FAs. (C) 2002 Elsevier Science Inc. Аннотация: This work considers antioxidant properties of natural detoxifying agents-humic substances (HS) in solutions of model inorganic and organic compounds of oxidative nature-complex salt K-3[Fe(CN)(6)] and 1,4-benzoquinone. Bioluminescent system of coupled enzymatic reactions catalyzed by NAD(P) H:FMN-oxidoreductase and bacterial luciferase was used as a bioassay in vitro to monitor toxicity of the oxidizer solutions. Toxicities of general and oxidative types were evaluated using bioluminescent kinetic parameters-bioluminescence intensity and induction period, respectively. Antioxidant activity of HS was attributed to their ability to decrease both general and oxidative toxicities; the HS antioxidant efficiency was characterized with detoxification coefficients D-GT and D-OxT, respectively. Dependencies of D-GT and D-OxT on HS concentration and time of preliminary incubation of the oxidizers with HS were demonstrated. The optimal conditions for detoxification of the oxidizers were >20-min incubation time and 0.5x10(-4) to 2x10(-4) M of HS concentration. The present study promotes application of the enzymatic luminescent bioassay to monitor toxicity of pollutants of oxidative nature in environmental and waste waters in remediation procedures.
