Аннотация: The effect of aflatoxin B1 on growth and luminescence of marine luminous bacteria P. phosphoreum and recombinant E. coli Z905 cells was investigated. The bidirectional effect of aflatoxin B1 on the studied bacterial species was detected-an inhibition of luminescence in P. phosphoreum and its stimulation in E. coli. It was shown that aflatoxin B1 influences the cell luminescence in the freshly grown cultures and bacteria restored after lyophilization. It was detected that the effect of aflatoxin B1 was graded after interaction with the modified nanodiamond (MND) of detonation synthesis. After mycotoxin's treatment with MND, it does not cause significant changes in bacterial luminescence. The possibilities for the use of P. phosphoreum and E. coli bacteria in the bioluminescent monitoring of aflatoxin B1 and the use of MND for mycotoxin deactivation are discussed.

Аннотация: Growth and potential photosynthetic activity of phytoplankton passed through intestine of crucian carp (Carassius auratus) from a small Siberian reservoir were compared with those of phytoplankton taken the directly from the reservoir. The dominant phytoplankton species in the reservoir, Microcystis aeruginosa, showed a significant increase of growth after the passage. Subdominant Planktothrix agardhii also showed an increase in growth rate, while subdominants Anabaena flos-aquae and Aphanizomenon flos-aquae were not stimulated by the gut passage.

Аннотация: Long-chain omega-3 polyunsaturated fatty acids (LC-PUFA) essential for human nutrition are mostly obtained from wild-caught fish. To sustain the LC-PUFA supply from natural populations, one needs to know how environmental and intrinsic factors affect fish fatty acid (FA) profiles and contents. We studied seven Salmoniformes species from two arctic lakes. We aimed to estimate differences in the FA composition of total lipids and two major lipid classes, polar lipids (PL) and triacylglycerols (TAG), among the species and to evaluate LC-PUFA contents corresponding to PL and TAG in muscles. Fatty acid profiles of PL and TAG in all species were characterized by the prevalence of omega-3 LC-PUFA and C16-C18 monoenoic FA, respectively. Fish with similar feeding spectra were identified similarly in multivariate analyses of total lipids, TAG and PL, due to differences in levels of mostly the same FA. Thus, the suitability of both TAG and total lipids for the identification of the feeding spectra of fish was confirmed. All species had similar content of LC-PUFA esterified as PL, 1.9-3.5 mg g(-1), while the content of the TAG form strongly varied, from 0.9 to 9.8 mg g(-1). The LC-PUFA-rich fish species accumulated these valuable compounds predominately in the TAG form.

salmonid fishes of families Coregonidae and Salmonidae -- polyunsaturated -- fatty acids -- eicosapentaenoic acid (EPA) -- docosahexaenoic acid (DHA) -- embryogenesis
Аннотация: A hypothesis was advanced and grounded that the total content of eicosapentaenoic (EPA, 20:5n-3) and docosahexaenoic (DHA, 22:6n-3) acids in fish muscle tissue is associated with the species-specific (taxon-specific) duration of embryo development. A meta-analysis of the original and published data was performed using fishes of the families Coregonidae and Salmonidae as an example. Fishes with longer embryo development times, which are observed at lower temperatures, were found to have significantly higher EPA + DHA contents in muscles as compared with the species that belong to the same families but have shorter embryo development times. This association was explained by the fact that an embryo forms more cells per unit tissue volume at lower temperatures, which requires a greater specific amount of cell membranes and, therefore, greater amounts of EPA and DHA to produce them.

Modeling -- Diet -- Dish -- Ingredient -- Nutrient -- Imbalance
Аннотация: The capability of "dish" and "ingredient in dish" modeling to reduce the number of nutrition imbalances in bioregenerative life support diet was compared. Masses of dishes were assumed to be the independent variables in the 'dish' model, while in the 'ingredient in dish' model the independent variables were the total masses of the ingredients in a one-day menu and masses of ingredients in the dishes. The objective function in both models was minimization of discrepancy between the calculated nutrition intakes and the daily nutrition requirements of NASA for long duration space missions. Comparing of two models was carried out for the case of a one-day diet containing 12 dishes and 32 ingredients. It was established that the diet simulation by 'dish' model brings 6 nutrition imbalances. The use of the 'ingredient in dish' modeling has helped to reduce the number of nutrition imbalances to 3, namely, an excess of iron, vitamin A and saturated fat. Obstacles to the fulfillment of all nutrition requirements were the nomenclature and masses of ingredients in the dish recipes.

Ciliates -- Meromictic lakes -- Chemocline -- Meromixis breakdown
Аннотация: The study deals with the vertical distribution and seasonal dynamics of planktonic ciliates in the pelagic and littoral zones of a saline meromictic Lake Shira. Fourteen species of free-living ciliates have been found in the Lake, seven of them inhabiting the pelagic zone. The richness of ciliates is higher both in terms of the number of species and biomass in the littoral zone compared to the pelagic zone. Although the ciliate species diversity is low in the pelagic zone, in certain seasons, the biomass of some of the species may reach considerable values, up to 3.4 g m(-2) in the water column. The biomass of ciliates in Lake Shira is generally comparable to the average values for other mesotrophic lakes. The abundance and composition of ciliate populations in Lake Shira vary considerably with depth. Changes in the Lake ecosystem caused by meromixis breakdown in 2015-2016, which induced considerable variations in many Lake components, affected the vertical distribution of planktonic ciliates. However, the annual average biomass of the species that inhabited the Lake before meromixis breakdown remained unchanged. The total ciliate biomass increased due to the presence of the new species. Bioluminescent biosensor -- Enzyme inhibition-based assay -- Complex -- matrices -- Pesticides -- Heavy metals
Аннотация: The presence of potentially toxic xenobiotics in complex matrices has become rather the rule than the exception. Therefore, there is a need for highly sensitive inexpensive techniques for analyzing environmental and food matrices for toxicants. Enzymes are selectively sensitive to various toxic compounds, and, thus, they can be used as the basis for detection of contaminants in complex matrices. There are, however, a number of difficulties associated with the analysis of complex matrices using enzyme assays, including the necessity to take into account properties and effects of the natural components of the test media for accurate interpretation of results. The present study describes the six-stage procedure for designing new enzyme sensors intended for assessing the quality of complex matrices. This procedure should be followed both to achieve the highest possible sensitivity of the biosensor to potentially toxic substances and to minimize the effect of the uncontaminated components of complex mixtures on the activity of the biosensor. The proposed strategy has been tested in designing a bioluminescent biosensor for integrated rapid assessment of the safety of fruits and vegetables. The biosensor is based on the coupled enzyme system NAD(P)H:FMN-oxidoreductase and luciferase as the biorecognition element. The study describes methods and techniques for attaining the desired result in each stage. The proposed six-stage procedure for designing bioluminescent enzyme biosensors can be used to design the enzymatic biosensors based on other enzymes.
