Аннотация:
Recombinant apoobelin and recombinant luciferase are separated from bacterial cells of Escherichia coli with the use of detonation nanodiamonds. The application of nanodiamonds has a number of points in its favor, namely, (i) simplifies the procedures for purifying the proteins, (ii) decreases the time of their separation to 30-40 min, (iii) eliminates the necessity of using special chromatographic equipment, and (iv) makes it possible to prepare high-purity apoobelin and luciferase materials with protein yields of 35-45 and 45-60%, respectively. The possible mechanisms of interaction of protein molecules and nanodiamond particles are analyzed. (C) 2004 MAIK "Nauka / Interperiodica".

Аннотация:
The electronic structure and total energy of various isomeric forms of coelenterazine and coelenteramide have been calculated by quantum chemistry methods both in the single-electron approximation and taking into account correlation effects. It has been shown that the inclusion of electron correlations makes it possible to obtain the structure of the coelenteramide close to the experimentally determined structure, as well as to choose the structure of the coelenterazine CLZ(1H) as the most probable isomeric form.

Са2+-регулируемый фотопротеин обелин определяет свечение морского гидроида Obelia longissima. Биолюминесценция инициируется кальцием и возникает вследствие окислительного декарбоксилирования связанного с белком субстрата целентеразина. Люцифераза светящегося морского коралла Renilla muelleri (RM) также использует целентеразин в качестве субстрата. Однако в биолюминесценцию этих животных in vivo вовлечено три белка: люцифераза, зеленый флуоресцентный белок и Са2+-регулируемый целентеразин-связывающий белок (coelenterazine-binding protein (СВР)). Фактически, ?субстратом? люциферазы RM в биолюминесцентной реакции in vivo является CBP, содержащий одну молекулу прочно связанного целентеразина. Целентеразин становится доступным для реакции с люциферазой и кислорода только после связывания с CBP ионов кальция. В отличие от Са2+-регулируемых фотопротеинов в молекуле CBP не происходит активации кислородом молекулы целентеразина. В работе с помощью квантово-химических методов исследовано поведение субстратов в данных белках. Показано, что целентеразин может образовывать различные таутомерные формы - CLZ(2H) и CLZ(7H). Исследован механизм образования 2-гидроперокси-целентеразина. Согласно полученным данным эти белки используют для реакции различные формы субстратов: в обелине субстрат находится в форме CLZ(2H), из которой и происходит образование гидроперекиси. В RM целентеразин находится в форме CLZ(7H), и поэтому активации кислородом в CBP не происходит.
The Са2+-regulated photoprotein
obelin
determines the luminescence of the marine hydroid Obelia longissima. Bioluminescence is initiated by calcium and appears as a result of the oxidative decarboxylation related to the coelenterazine substrate. The luciferase of the luminescent marine coral Renilla muelleri (RM) also uses coelenterazine as a substrate. However, three proteins are involved in the in vivo bioluminescence of these animals: luciferase, green fluorescent protein, and Са2+-regulated coelenterazine-binding protein (СВР). In fact, CBP that contains one strongly bound coelenterazine molecule is the RM luciferase substrate in the in vivo bioluminescent reaction. Coelenterazine becomes available for oxygen and the reaction with luciferase only after binding CBP with calcium ions. Unlike Са2+-regulated photoproteins, the coelenterazine molecule is not activated by oxygen in the CBP molecule. In this work, by means of quantum chemical methods the behavior of substrates in these proteins is analyzed. It is shown that coelenterazine can form different tautomers: CLZ(2H) and CLZ(7H). The formation of 2-hydroperoxy-coelenterazine is studied. According to the obtained data, these proteins use different forms of the substrates for the reaction. In

Coelenteramide-containing fluorescent proteins are products of bioluminescent reactions of marine coelenterates. They are called ‘discharged photoproteins’. Their light-induced fluorescence spectra are variable, depending considerably on external conditions. Current work studies a dependence of light-induced fluorescence spectra of discharged photoproteins
obelin
, aequorin, and clytin on excitation energy. It was demonstrated that photoexcitation to the upper electron-excited states (260–300 nm) of the discharged photoproteins initiates a fluorescence peak in the near UV region, in addition to the blue-green emission. To characterize the UV fluorescence, the light-induced fluorescence spectra of coelenteramide (CLM), fluorophore of the discharged photoproteins, were studied in methanol solution. Similar to photoproteins, the CLM spectra depended on photoexcitation energy; the additional peak (330 nm) in the near UV region was observed in CLM fluorescence at higher excitation energy (260–300 nm). Quantum chemical calculations by time depending method with B3LYP/cc-pVDZ showed that the conjugated pyrazine-phenolic fragment and benzene moiety of CLM molecule are responsible for the additional UV fluorescence peak. Quantum yields of CLM fluorescence in methanol were 0.028 ± 0.005 at 270–340 nm photoexcitation. A conclusion was made that the UV emission of CLM might contribute to the UV fluorescence of the discharged photoproteins. The study develops knowledge on internal energy transfer in biological structures – complexes of proteins with low-weight aromatic molecules. © 2016 Elsevier B.V.

Bioluminescent solid-phase analysis was proposed to monitor the selection process and to determine binding characteristics of the aptamer-target complexes during design and development of the specific aptamers. The assay involves Ca2+-regulated photoprotein
obelin
as a simple, sensitive and fast reporter. Applicability and the prospects of the approach were exemplified by identification of DNA aptamers to cardiac troponin I, a highly specific early biomarker for acute myocardial infarction. Two structurally different aptamers specific to various epitopes of troponin I were obtained and then tested in a model bioluminescent assay.
