Аннотация: The article presents the results of investigation of antitumor properties of platinum–arabinogalactan complex. We showed the ability of the complex to inhibit the growth of Ehrlich ascites tumor cells. It is found that the distribution of the platinum–arabinogalactan complex is not specific only for tumor cells in mice. The complex was found in all tissues and organs examined (ascites cells, embryonic cells, kidney, and liver). The mechanism of action of the arabinogalactan–platinum complex may be similar to cisplatin as the complex is able to accumulate in tumor cells. © 2016, Pleiades Publishing, Ltd.

Аннотация: Nanotechnologies involving physical methods of tumor destruction using functional oligonucleotides are promising for targeted cancer therapy. Our study presents magnetodynamic therapy for selective elimination of tumor cells in vivo using DNA aptamer-functionalized magnetic nanoparticles exposed to a low frequency alternating magnetic field. We developed an enhanced targeting approach of cancer cells with aptamers and arabinogalactan. Aptamers to fibronectin (AS-14) and heat shock cognate 71 kDa protein (AS-42) facilitated the delivery of the nanoparticles to Ehrlich carcinoma cells, and arabinogalactan (AG) promoted internalization through asialoglycoprotein receptors. Specific delivery of the aptamer-modified FeAG nanoparticles to the tumor site was confirmed by magnetic resonance imaging (MRI). After the following treatment with a low frequency alternating magnetic field, AS-FeAG caused cancer cell death in vitro and tumor reduction in vivo. Histological analyses showed mechanical disruption of tumor tissues, total necrosis, cell lysis, and disruption of the extracellular matrix. The enhanced targeted magnetic theranostics with the aptamer conjugated superparamagnetic ferroarabinogalactans opens up a new venue for making biocompatible contrasting agents for MRI imaging and performing non-invasive anti-cancer therapies with a deep penetrated magnetic field.

Аннотация: The antioxidant properties of selenium nanoparticles coated with arabinogalactan have been studied. The nanoparticles were characterized by transmission electron microscopy and infrared spectroscopy. Arabinogalactan-coated selenium nanoparticles were incubated together with Ehrlich ascitic carcinoma cells and then exposed to microwave radiation. The antioxidant properties of selenium nanoparticles were evaluated using chemiluminescent analysis. The viability of Ehrlich ascitic carcinoma cells under microwave irradiation and incubation with selenium nanoparticles was assessed by flow cytometry. Under conditions of exposure to experimental tumor cells of microwave radiation, the concentration of free radicals in the cells did not change. The effect of arabinogalactan-coated selenium nanoparticles on tumor cells is expressed as an increase in the delay time for the activation of the antioxidant system.

Аннотация: The effect of magnetic nanoparticles coated with arabinogalactan on the viability of Ehrlich ascites carcinoma (EAC) cells was studied. The nanoparticles were studied by transmission electron microscopy, Mossbauer spectroscopy, IR spectroscopy, and ferromagnetic resonance. A correlation between the proportion of dead EAC cells in suspension and the intensity of the EPR signal of dinitrosyl iron complexes was found. This result may be due to the presence of NO molecules.

Аннотация: In this study, nanoparticles of initial synthetic ferrihydrite have been coated with arabinogalactan. The synthesized series of samples with different degrees of coverage of particles has been characterized by X-ray photoelectron spectroscopy, Mossbauer spectroscopy, transmission electron microscopy and magnetometry. The superparamagnetic blocking temperature decreases monotonically with an increase in the degree of coverage of ferrihydrite particles, which is unambiguously related to the different role of the interparticle magnetic interactions in the investigated powder systems. Analysis of the field dependence of the blocking temperature within the random anisotropy model has shown that an increase in the degree of coverage of ferrihydrite particles leads to a decrease in the size of a cluster in which the behaviors of the nanoparticle magnetic moments are correlated. The results obtained have shown the possibility of effective control of the strength of magnetic interparticle interactions in powder ferrihydrite systems by coating nanoparticles with arabinogalactan.

Аннотация: In this work, we investigated the ferromagnetic resonance spectra of metal/carbon composite coatings. FeC and NiC coatings were synthesized by electroless deposition using polysaccharide arabinogalactan. An analysis of the angular dependences of the resonance field showed that the coatings consist of three magnetic phases separated by a nonmagnetic phase of carbon.

The particles consist of an anisomeric magnetite core coated with arabinogalactan and are functionalized with cameras for As-14 ascites cells (Ehrlich carcinoma). The binding of ascitic Ehrlich carcinoma cells to magnetic nanoparticles was evaluated using fluorescence microscopy.
