Аннотация: We studied the feasibility of using a short-term culture of monocytes, isolated from peripheral donor blood, to assess the biological activity of different types of bionanomaterials (BNM): biodegradable polimeric particles, fiber and film substrates of micro- and nano-dimensions, fullerenes (F) and nanodiamonds (ND), which are either currently in use and/or potentially applicable in medicine. Additionally, the effect of creating a protein corona on ND and F particles was investigated. The cellular reduction of (4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) is a well-established tool for assessing the viability/metabolic activity of cells. The scanning electron microscopy assay can detect fine changes in cell morphology. In the present study BNM have been shown to affect; in a size, chemical composition and morphological characteristics-dependent manner, the ability of monocytes to reduce MTT as well as their morphology. Moreover, the specific effects of ND and F on MTT reduction and cell morphology were exhibited in a dose-dependent manner and sensitive to the formation of surface protein corona. Our results suggest that short-term culture of monocytes is a sensitive model system for assessing the biological effects of BMPs in vitro. © 2016 Elsevier Ltd

Аннотация: Изучены морфофункциональные особенности моноцитов в условиях кратковременного культивирования in vitro на носителях из биополимера полигидроксиалканоата (ПГА) в присутствии наноалмазов (НА) и фуллеренов (Ф). Показано, что НА и Ф в исследованных концентрациях не влияли на жизнеспособность моноцитов в кратковременной культуре. Соотношение морфологических классов моноцитов в присутствии НА и Ф существенно отличалось от контрольного варианта культивирования. Влияние НА и Ф характеризовалось дозовой зависимостью. Воздействие наночастиц на культуру моноцитов значительно изменялось при формировании белковой короны на поверхности НА и Ф, что приводило к возникновению новых количественных соотношений между морфологическими классами моноцитов в культуре in vitro. Полученные результаты позволяют рассматривать кратковременную культуру моноцитов in vitro как адекватную модельную систему для оценки влияния новых материалов на процессы дифференцировки субпопуляций моноцитов.
The morphological and functional features of the monocytes in a short-term in vitro cultivation on substrates of biopolymer polyhydroxyalkanoate (PHA) in the presence of nanodiamonds (ND) and fullerene (F) were studied. It has been shown that ND and F in the studied concentrations did not affect the viability of monocytes in the short-term culture. The ratio of morphological classes in the presence of ND and F signi?cantly different from the control group of cultivation. Effects of NA and F were characterized by dose-dependent. The formation of a protein corona on the surface of NA and F led to signi?cant changes in the effects of nanoparticles and the formation of new quantitative relationships between morphological classes of monocytes in culture in vitro. The results allow to consider a short-term culture of monocytes in vitro as a model system for adequate assessment of the impact of new materials on the processes of differentiation of monocyte subpopulations.

Аннотация: An "aluminum oxide film-adhesive layer-nanodiamond-luciferase" supramolecular structure is prepared on a flat plate. It is demonstrated that, in this structure, the enzyme retains the catalytic activity. The structure prepared can be treated as a luminescent biochip prototype for use in bioluminescent analysis. Аннотация: Experimental data have been presented on the influence of silver on the viscosity and thermal conductivity of a dispersion of diamond nanoparticles. A stable dispersion (5 wt %) of detonation nanodiamond particles has been used in the experiments. Silver ions have been introduced electrolytically into the dispersion of diamond nanoparticles. Silver concentration was not higher than 0.05 wt %. It has been shown that the introduction of silver ions significantly affects the thermal conductivity and viscosity of the dispersion.

Аннотация: Phenolic compounds are common environmental pollutants, which are used in many industries and contaminate water environment due to industrial wastewater discharge. Presently, there is an increasing demand for the development and improvement of inexpensive, easy-to-use analytical tools for in-situ detection of phenolic compounds. In this work, we investigate the effect of adsorption of Cu2+ and Fe2+ ions on the catalytic activity of a composite material based on alumina nanofibers (ANF) and detonation nanodiamonds (DND) in the co-oxidation of phenols with 4-aminoantipyrine in the presence of hydrogen peroxide. We have found more than two-fold increase of the catalytic activity for ANF + DND + Cu composite, while the activity of ANF + DND + Fe composite is found to decrease by several times in comparison with the original ANF + DND material. The results of FTIR analysis indicate that the adsorption of iron ions occurs with the formation of hydroxide surface groups and hydrogen bonds, which apparently block their catalytic activity in the Fenton redox cycle. The higher catalytic performance of AND + DND composite functionalized with copper ions makes it possible to detect two times lower concentrations of analytes (phenol and 4-chlorophenol) in comparison with the original composite. It is shown that the AND + DND + Cu composite provides a linear yield of the co-oxidation reaction product in a wide range of analyte concentrations (0.25–100 μM for phenol and 0.5–25 μM for 4-chlorophenol). Model experiments demonstrate the applicability of copper-functionalized composite as a reusable sensor for the determination of phenol in aqueous samples.

Аннотация: This article presents experimental data on the coefficients of viscosity and thermal and electrical conductivities, as well as the absorption spectra, of suspensions of ultradispersed diamonds. Suspensions of UDA-S and UDP-A detonation nanodiamond particles, as well as UDP-AG diamond–graphite powder, are used in the experiments. The concentration of nanodiamonds in distilled water is varied within a range of 0.5–5 wt %. It is shown that the technology of purification diamond powders from side products of their synthesis substantially affects the physicochemical properties of aqueous suspensions of these powders.

Аннотация: Recombinant apoobelin and recombinant luciferase are separated from bacterial cells of Escherichia coli with the use of detonation nanodiamonds. The application of nanodiamonds has a number of points in its favor, namely, (i) simplifies the procedures for purifying the proteins, (ii) decreases the time of their separation to 30-40 min, (iii) eliminates the necessity of using special chromatographic equipment, and (iv) makes it possible to prepare high-purity apoobelin and luciferase materials with protein yields of 35-45 and 45-60%, respectively. The possible mechanisms of interaction of protein molecules and nanodiamond particles are analyzed. (C) 2004 MAIK "Nauka / Interperiodica".
