Аннотация: Controlled drug delivery is one of the frontier areas of science, which uses an interdisciplinary approach. The delivery systems offer numerous advantages over conventional dosage forms, such as improved efficacy and patient compatibility, reduced toxicity, and ease of use. Such systems often use micro- and nanoparticles as carriers for drugs, the prolonged effect of which is achieved due to the controlled slow release of the encapsulated drug. This study investigated the effects of encapsulation of various antiseptics (brilliant green, miramistin, and furacilin) and the chemical composition of the polymer on the yield, structure, size, drug release kinetics, and antibacterial activity of microparticles produced from resorbable polyesters of microbiological origin, polyhydroxyalkanoates. Microparticles with a 5.6–94.8 μm diameter were produced. The form of the active substance molecule has been found to be the most significant factor affecting the characteristics of polyhydroxyalkanoate microparticles. The surface structure of particles is rather determined by the chemical composition of the polymer, and the release kinetics to the model medium depends on the encapsulated drug. Microparticles based on PHAs loaded with brilliant green and furacilin showed antibacterial effects in S. aureus and E. coli cultures. The study demonstrated that microparticles with antiseptics encapsulated in them have potential as prolonged drug delivery systems and are of interest for further research.Технология контролируемой доставки лекарств является одной из передовых областей науки, включающей междисциплинарный научный подход. Эти системы доставки обладают многочисленными преимуществами по сравнению с обычными лекарственными формами, такими как повышенная эффективность и биосовместимость, сниженная токсичность и простота использования. В таких системах в качестве носителей лекарственных средств часто используют микро- и наночастицы, пролонгированное действие которых достигается за счет контролируемого медленного высвобождения инкапсулированного лекарственного средства. В данном исследовании изучалось влияние включения различных антисептиков (бриллиантовый зелёный, мирамистин и фурацилин), а также химического состава полимера на выход, структуру, размер, кинетику высвобождения лекарственных средств и антибактериальную активность микрочастиц, полученных из резорбируемых полиэфиров микробиологического происхождения, полигидроксиалканоатов. Было получено семейство микрочастиц диаметром 5,6–94,8 мкм. Установлено, что наиболее значимым фактором, влияющим на характеристики микрочастиц полигидроксиалканоатов, является форма молекулы активного вещества. Структура поверхности частиц больше зависит от химического состава полимера, а кинетики выхода в модельную среду – от инкапсулированного лекарства. Микрочастицы на основе ПГА с депонированными бриллиантовым зеленым и фурацилином показали антибактериальное действие в культурах S. aureus и E. coli. Результаты продемонстрировали, что микрочастицы с депонированными антисептиками обладают потенциалом в качестве систем пролонгированной доставки и представляют интерес для дальнейших исследований.

Аннотация: Biodegradable polyhydroxyalkanoates, biopolymers of microbiological origin, formed by 3- and 4-hydroxybutyrate monomers P(3HB-co-4HB), were used to obtain nanomembranes loaded with drugs as cell carriers by electrospinning. Resorbable non-woven membranes from P(3HB-co-4HB) loaded with ceftazidime, doripinem, and actovegin have been obtained. The loading of membranes with drugs differently affected the size of fibers and the structure of membranes, and in all cases increased the hydrophilicity of the surface. The release of drugs in vitro was gradual, which corresponded to the Higuchi and Korsmeyer-Peppas models. Antibiotic-loaded membranes showed antibacterial activity against S. aureus and E. coli, in which growth inhibition zones were 41.7 ± 1.1 and 38.6 ± 1.7 mm for ceftazidime and doripinem, respectively. The study of the biological activity of membranes in the NIH 3T3 mouse fibroblast culture based on the results of DAPI and FITC staining of cells, as well as the MTT test, did not reveal a negative effect despite the presence of antibiotics in them. Samples containing actovegin exhibit a stimulating effect on fibroblasts. Biodegradable polyhydroxyalkanoates formed by 3-hydroxybutyrate and 4-hydroxybutyrate monomers provide electrospinning non-woven membranes suitable for long-term delivery of drugs and cultivation of eukaryotic cells, and are promising for the treatment of wound defects complicated by infection.

Аннотация: The properties, features of thermal behavior and crystallization of copolymers containing various types of valerate monomers were studied depending on the set and ratio of monomers. We synthesized and studied the properties of three-component copolymers containing unusual monomers 4-hydroxyvalerate (4HV) and 3-hydroxy-4-methylvalerate (3H4MV), in addition to the usual 3-hydroxybutyrate (3HB) and 3-hydroxyvalerate (3HV) monomers. The results showed that P(3HB-co-3HV-co-4HV) and P(3HB-co-3HV-co-3H4MV) terpolymers tended to increase thermal stability, especially for methylated samples, including an increase in the gap between melting point (Tmelt) and thermal degradation temperature (Tdegr), an increase in the melting point and glass transition temperature, as well as a lower degree of crystallinity (40–46%) compared with P(3HB-co-3HV) (58–66%). The copolymer crystallization kinetics depended on the set and ratio of monomers. For terpolymers during exothermic crystallization, higher rates of spherulite formation (Gmax) were registered, reaching, depending on the ratio of monomers, 1.6–2.0 µm/min, which was several times higher than the Gmax index (0.52 µm/min) for the P(3HB-co-3HV) copolymer. The revealed differences in the thermal properties and crystallization kinetics of terpolymers indicate that they are promising polymers for processing into high quality products from melts.

Аннотация: Biodegradation of microbial linear polymers of hydroxyalkanoic acids (polyhydroxyalkanoates, PHAs) by soil microbial communities of different structures has been studied during two field seasons in different weather conditions. This process was shown to be influenced by the polymer chemical composition, temperature, humidity, and the microbial soil component. The PHA degradation was accompanied by a decrease in the polymer molecular weight and an increase in the degree of crystallinity, indicating the preferential destruction of the amorphous phase compared to the crystalline one. The quantity of the true PHA destructors developing at the surface of the polymer samples was lower than the quantity of accompanying bacteria. The dominant PHA degrading microorganisms under the test conditions were identified as bacteria of the genera Variovorax, Stenotrophomonas, Acinetobacter, Pseudomonas, Bacillus, and Xanthomonas and as micromycetes from Penicillium, Paecilomyces, Acremonium, Verticillium, and Zygosporium.

Аннотация: BACKGROUND: Polyhydroxyalkanoates (PHA) containing 3-hydroxy-4-methylvalerate (3H4MV) have better processing properties than other PHA types and were not prone to ageing. The main challenge in the production of these polymers is to simultaneously achieve high cell biomass, high polymer content, and high molar fraction of 3H4MV. The aim of this study was to investigate the production of poly(3-hydroxybutyrate-co-3-hydroxyvalerate-co-3-hydroxy-4-methylvalerate) [P(3HB-co-3 HV-co-3H4MV)] by Cupriavidus eutrophus B-10646, synthesize polymers with different contents of 3H4MV and investigate their properties. RESULTS: It was shown that 4-methylvalerate (4MV, the precursor substrate) concentration in the culture medium must not be higher than 1 g L−1, and that 4MV must be carefully dosed, with at least 10–12 h intervals between supplementations. A series of polymers with molar fractions of 3H4MV between 2.7 and 11.3 mol% was synthesized. Degrees of crystallinity, molecular weight characteristics, and thermal properties of the polymers have been investigated as dependent on proportions of monomers. CONCLUSION: The content of 3H4MV depends on the concentration of 4MV and conditions of carbon nutrition. The molar fraction of 3H4MV had a strong effect on the crystalline to amorphous region ratio. PHA containing 3H4MV specimens synthesized in this study had a degree of crystallinity of below 50%, which remained unchanged for 2.5 years.

Аннотация: The study addresses the growth of the wild-type strain Cupriavidus necator B-10646 and the synthesis of sulfur-containing polyhydroxyalkanoates (PHA) by this strain on media containing fructose and three different precursors (3-mercaptopropionic acid, 3′,3′-dithiodipropionic acid and 3′,3′-thiodipropionic acid). By varying the concentration and number of doses of the precursors added into the bacterial culture, it was possible to find conditions that ensure the formation of 3-mercaptopropionate (3MP) monomers from the precursors and their incorporation into the C-chain of poly(3-hydroxybutyrate). A series of P(3HB-co-3MP) copolymer samples with different content of 3MP monomers (from 2.04 to 39.0 mol.%) were synthesized and the physicochemical properties were studied. The effect of 3MP monomers is manifested in a certain decrease in the molecular weight of the samples and an increase in polydispersity. Temperature changes are manifested in the appearance of two peaks in the melting region with different intervals regardless of the 3MP content. The studied P(3HB-co-3MP) samples, regardless of the content of 3MP monomers, are characterized by equalization of the ratio of the amorphous and crystalline phases and have a close degree of crystallinity with a minimum of 42%, = and a maximum of 54%.

Аннотация: Synthesis of polyhydroxyalkanoates (PHAs) by a new strain of Cupriavidus - Cupriavidus eutrophus B-10646 - was investigated under autotrophic growth conditions. Under chemostat, at the specific flow rate D=0.1h-1, on sole carbon substrate (CO2), with nitrogen, sulfur, phosphorus, potassium, and manganese used as growth limiting elements, the highest poly(3-hydroxybutyrate) [P(3HB)] yields were obtained under nitrogen deficiency. In batch autotrophic culture, in the fermenter with oxygen mass transfer coefficient 0.460h-1, P(3HB) yields reached 85% of dry cell weight (DCW) and DCW reached 50g/l. Concentrations of supplementary PHA precursor substrates (valerate, hexanoate, ?-butyrolactone) and culture conditions were varied to produce, for the first time under autotrophic growth conditions, PHA ter- and tetra-polymers with widely varying major fractions of 3-hydroxybutyrate, 4-hydroxybutyrate, 3-hydroxyvalerate, and 3-hydroxyhexanoate monomer units. Investigation of the high-purity PHA specimens showed significant differences in their physicochemical and physicomechanical properties. В© 2013 Elsevier Ltd.

Аннотация: Films from biodegradable poly-3-hydroxybutyrate are treated with chemical reagents to improve their hydrophilicity and biocompatibility. Two approaches are tested: a single treatment with alkali, acids, oxidizing or reducing agents, and a step-by step treatment of the alkali pre-activated surface of polymer films with bromine water and amino-compounds (ammonia or triethylamine). The maximal level of hydrophilicity (the lowest water contact angle and the highest polar component of the surface free energy) is registered after a single treatment with NaOH and after the step-by-step treatment. These samples also showed the best adhesion of mouse fibroblasts of NIH 3T3 line on the film surface. So, the proposed methods can be used to enhance hydropilicity and biocompatibility of biopolymer surface.

Аннотация: IIn this study, electrospinning was used to prepare ultrafine fibers from PHAs with different chemical compositions: P(3HB) and copolymers: P(3HB-co-4HB), P(3HB-co-3HV), and P(3HB-co-3HHx). The main process parameters that influence ultrafine fiber diameter and properties (polymer concentration, solution feeding rate, working distance, and applied voltage) have been investigated and their effects evaluated. The study revealed electrospinning parameters for the production of high-quality ultrafine fibers and determined which parameters should be varied to tailor the properties of the products. This study is the first to compare biological and physical-mechanical parameters of PHAs with different chemical compositions as dependent upon the fractions of monomers constituting the polymers and ultrafine fiber orientation. Mechanical strength of aligned ultrafine fibers prepared from different PHAs is higher than that of randomly oriented ones; no significant effect of ultrafine fiber orientation on surface properties has been found. None of the fibrous scaffolds produced by electrospinning from PHAs had any adverse effects on attachment, growth, and viability of NIH 3T3 mouse fibroblast cells, and all of them were found to be suitable for tissue engineering applications.
